Objective: To explore the effect and significance of Toll-likereceptors (TLR2, TLR3) on C3a and C5a in patients with wet-AMD.Methods: Peripheral blood samples were collected from15patientswith wet-AMD and as well as15subjects without wet-AMD as control.Peripheral blood mononuclear cells were then isolated using gradientcentrifugation. We divided the cells into7groups, namely blank group,C3a group, C5a group, TLR2on C3a group, TLR2on C5a group, TLR3onC3a group and TLR3on C5a group. Stimulations were added to each group.The supernatant were collected respectively and were tested forconcentration of AMD–related cytokines, such as IL-1β, IL-6, IL8, IL-17, MCP-1, VEGF, applying ELISA technology.Results: The Toll-like receptors on complement system from bothpatients with wet-AMD and control group demonstrated higherconcentrations of IL-1β, IL-6, IL-8, MCP–1, compared with C3a orC5a group; patients with wet–AMD groups showed higher concentrationsof IL-1β, IL-6, IL–8and MCP-1compared with the control groupsrespectively. Conclusion: The effects of Toll-like receptors on the complementsystem appears to promote the development of the wet-AMD.
