Objective:To detect the expressions of vascular endothelial growth factor(VEGF), matrixmetalloproteinase-2(MMP-2) and matrix metalloproteinase9(MMP-9) in endometrialpolyps and malignant endometrial polyps, and to probe the correlation for expressionof VEGF, MMP-2and MMP-9and the mechanism underlying pathogenesy ofendometrial polyps.Methods:Specimen collection:60patients with endometrial polyps tissue samples inAffiliated hospital of XiangNan University from December2009to December2012as the observation group, which includes30cases of atypical hyperplasia ofendometrial polyps tissue samples and30cases of simple hyperplasia endometriumpolyp tissue samples. Then chose the same period30cases of the endometrial polypsor polyp pedicle of endometrial tissue samples of as control group1, and chose30cases of normal endometrial tissue samples as control group2.Immunohistochemical staining: All example specimen after processing, and thenwith a microscope. to observe their protein expressions.Result judgment: The qualitative judgment according to the staining cells; adoptthe positive cell percentage of semi-quantitative method to determine the dyeingresults per milliliter of tissue containing two positive cells expressing decision basis, use the integral method to determine the degree of positive staining results of eachcase. All the results are independently evaluated by two pathologists.Statistical analysis: In this study, SPSS19.0statistical analysis method wasanalyzed statistical data. The groups sample is tested with X2; if data conform tonormality and homogeneity of variance, t-test is employed, otherwise, Wilcoxon rankstatistical analysis is employed. Values of P <0.05were considered statistically.Results:(1) The abnormal expression of VEGF in atypical hyperplasia of endometrialpolyps tissue samples and hyperplasia endometrium polyp tissue samples comparedwith the control group1and control2group. The difference was statisticalsignificance (P <0.05). In endometrial polyp group, VEGF positive percentage of thecomplexity of hyperplastic polyp group specimens is higher than that of simplehyperplasia polyps group, and the difference was statistical significance (x2=18.685,P <0.01), while difference between two groups in dyeing score (x2=2.685, P>2.685)were not obvious.(2) The expression of both MMP-2and MMP-9in observation group is obviouslyhigher than that of control1group and control group2, and the difference hasstatistical significance (P <0.05). In hyperplastic polyp group of specimens ofMMP-2and MMP-9positive percentage are91.2%(27/30) and88.2%(26/30), whichare significantly higher than that of simple hyperplasia polyps group as45.2%(14/30),40.7%(12/30). The dyeing score difference is statistically significant (x2=18.685, P <0.01; x2=21.243, P <0.05).(3) The expression of VEGF, MMP-2and MMP-9in hyperplasia endometrialpolyps group were positive correlation(F>0.05).Conclusion:(1) VEGF, MMP-2and MMP-9are over-expression in endometrial polyps. (2) The expressions of VEGF, MMP-2and MMP-9in complex hyperplasiaendometrial polyps are higher than the simple hyperplasia.(3) The expression of VEGF, MMP-2and MMP-9are positive correlation inendometrial polyps. |
