Objectives:The objectives of present work was to investigate the effects of dezocine onserum testosterone、follicle-stimulating hormone and luteinizing hormone levels inmale mice;the histopathological changes and the expressions of Bax and Bcl-2ofmouse testis.Methods:Sixty C57mice of six-week age and18-20g weight were divided into fivegroups randomly(n=12): control group,20mg/kg dose of morphine group,15mg/kg(low-dose) group、20mg/kg(middle-dose) group、25mg/kg (high-dose) groupof dezocine. The above treatments were intraperitoneally administered for21daysafter adaptive breeding for a week. The general condition was observed immediatelyand6hours after injection.The mice were sacrificed after experiment and the bloodand testis were taken immediately. The serum level of TSH,LH and TSH were testedby automatic biochemical analyzer. Testicular tissue pathological morphologicalchanges of mice were observed by HE staining. The expressions of Bax and Bcl-2intestis were measured by immunohistochemistry.Results:1. The behavior of the control group is normal. The mice of morphine group anddezocine groups were excited immediately after the drug injection.The behavior of themice returned to normal6hours later. 2.The mouse testicular pathology indicates that the seminiferous tubules of micein control group are intact, epithelial cell layers are normal, and there are lots ofmature sperm in the seminiferous tubule. The basement membrane of testis tissue inmorphine group are imcomplete. Epithelial cell layers are significantly reduced anddisorderly arranged. The spermatogenic epithelium of testis in dezocine group areunintegral.Epithelial cell layers disordered arrangement, with spermatogoniumdetachment or vacuolar degeneration. The primary spermatocyte, the secondaryspermatocyte, the spermatoblast and sperm in the seminiferous tubule are depressed,spermatocyte degenerated, sperm shape are abnormal in the dezoine groups. Thedestruction of seminiferous tubule from the dezocine groups is dose-dependent.3.The expression of Bax is significantly higher and the expression of Bcl-2issignificantly lower in morphine group and dezocine groups than that of controlgroup(p<0.05); the expression of Bax and Bcl-2are statistical significance amongD1,D2and D3group(p<0.05). Compared with morphine group, the expression of Baxis higher in D3group(p<0.05), while the expression of Bcl-2is lower(p<0.05).4.Compared with control group, the serum level of FSH,T and LH aresignificantly lower in high-dose dezocine group(p<0.05).The serum level of FSH,Tand LH in high-dose dezocine group are significantly lower than morphinegroup(p<0.05).Conclusion:The result show that administration of dezocine could induce mouse excited,suppress the level of plasma testosterone, follicle stimulating hormone and luteinizi-ing hormone, damage testicular structure, lead to the expression of Bax and Bcl-2protein imbalance, and affect male fertility. Therefore, it should be take a goodattention for the clinical applications.
