Objective:By observing GTW-induced expression of DOR rats serum level of IL-2and TNF-a,and to further explore the mechanism of action Bushenhuoxue square improve ovarian reserve,and provide scientific basis for the clinical treatment of DOR.Methods:Select healthy female SD rats are48,divided into six groups according to body weight(n=8):Control group(administered the same volume of saline),GTW model group(GTW50mg/kg/d),Liuweidihuang square group(GTW50mg+Liuweidihuang square7.50g crude/kg/d),Bushenhuoxue low dosage group(GTW50mg+Bushenhuoxue square3.25g crude drug/kg/d), Bushenhuoxue prescription dose group((GTW50mg+Bushenhuoxue square7.50g crude drug/kg/d), Bushenhuoxue high dose group(GTW50mg+Bushenhuoxue square15.00g crude drug/kg/d). Gavage for15days,1-4days after the administration of a combination of vaginal cytology smears,obviously will not appear in the estrous cycle in rats anesthetized with ether,to the femoral artery blood sacrificed by cervical dislocation and immediate removal of a large rat ovaries.Observed estrous cycle of the rats, body weight, ovarian mass, ovarian factor.Using radioimmunoassay method to detect serum levels of follicle stimulating hormone(FSH),estradiol(E2)and IL-2of rats;HE staining method detect morphological changes in ovarian tissue;Expression in rat ovarian tissue levels of TNF-α detected by immunohistochemistry through.Results:(1)Ovaries and ovarian mass coefficient model rats is lower than the control group;Model rats serum FSH significantly soaring, E2, IL-2decreased significantly(P values are p<0.001、p<0.001、p<0.01); Ovarian follicles in each period in the model rats were significantly reduced among different levels of follicle endoplasmic inflammatory cell infiltration and fibrosis;TNF-a in rats showed a lower expression,there is a significant difference with the control group (p<0.05).(2)Bushenhuoxue side of low, medium and high dose group and Liuweidihuang square group can improve GTW varying degrees rat ovary and ovarian mass coefficient, each group are able to significantly reduce the level of serum FSH of GTW rats, compare with model group showed significant differences(p<0.001). Bushenhuoxue prescription dose group and high-dose groups, and Liuweidihuang square group can elevate serum E2levels, compare with model group showed significant difference(P values are p<0.01、p<0.5、p<0.01); Bushenhuoxue prescription dose group and high-dose groups, and Liuweidihuang square group can be elevated serum levels of IL-2, with the model group are significantly different(both p<0.05); Bushenhuoxue prescription dose and high-dose groups granulosa cells and oocytes of TNF-a expression improve compared with the model group, there are significant differences between the model group (P<0.05),It show no significant differences between the control group.Conclusion:(1) Bushenhuoxue can fall tone serum FSH levels of GTW rats, increase serum levels of E2. This indicates Bushenhuoxue can improve ovarian reserve,increase serum levels of IL-2,antagonize the ovarian follicle damage,increase the expression of granulosa cells and oocytes of TNF-a,this is probably another prevention mechanism of improving DOR.(2) Bushenhuoxue and Liuweidihuang square could improve ovarian reserve and inhibition of follicle damage. But Bushenhuoxue party than Liuweidihuang square expression in granulosa cells and improve oocyte TNF-a aspects, this is consistent with the basis of previous studies. This reflects that Bushenhuoxue square can improve DOR through various mechanisms.
