Expression Of TGR5and NF-κB In Placental Tissue Of Intrahepatic Cholestasis Of Prengency

Objective To investigate the expression of TGR5and NF-κB inplacenta of intrahepatic cholestasis of pregnancy(ICP) and explore theeffect of TGR5and NF-κB in ICP．Methods Thirty-nine women with ICP who delivered from March2013to November2013in the First Affiliated Hospital of ChongqingMedical University were chosen as the study objects,among these,mildgroup(n=21)and severe group(n=18),while30cases of healthy gestationwomen(NG) who delivered through elective term cesarean sections ascontrol group(n=30). Immunohistochemistry was used to detect theexpression of TGR5and NF-κB.Results (1) TGR5and NF-κB expression was detected in placentalmacrophage and trophocyte of both groups,and was present in the cellmembrane and the cytoplasm.(2)In placental macrophage,TGR5expressionwas lower in ICP group than control group（Z=-4.237，P<0.05), and TGR5expression lower in severe ICP group than mild ICP group(Z=-2.351,P<0.05); NF-κB expression was higher in ICP group than control group （Z=-2.266，P<0.05), and NF-κB expression higher in severe ICP groupthan mild ICP group （Z=-3.017， P<0.05）.(3)In trophocyte,TGR5expression was also lower in ICP group than control group（Z=-2.397,P<0.05）, and TGR5expression lower in severe ICP group than mild ICPgroup();NF-κB expression was higher in ICP group than control group（Z=-3.207，P<0.05), and NF-κB expression higher in severe ICP groupthan mild ICP group（Z=-2.671，P<0.05）.(4) There was a negativecorrelation between TGR5and NF-κB in both macrophage andtrophocyte(r=-0.584,P<0.05;r=-0.424, P<0.05).Conclusions In ICP placental macrophage and trophocyte,a decreaseof TGR5is associated with increase of NF-κB. Furthermore, TGR5-NF-κBsignaling pathway maybe involve in pathophysiology of ICP.
