| Objective:Invasion and metastasis are two features of malignant tumor.To investigate the expression of MTA1, MMP-9and VEGF-C in gastric adenocarcinoma and the relationship between them and the clinicopathological features, and understand the role and correlation of these genes involved in tumor invasion and metastasis of the gastric adenocarcinoma development.Methods:We collecterd120cases of gastric adenocaircinoma specimens with complete clinicopathological information which diagnosed between Jun2012and May2013from our Hospital. We also selected30cases of normal gastric mucosa specimens from nonneoplastic epithelia of gastric adenocarcinoma as the control. According to the WHO(2004)pathological classification and UICC the TNM staging(2010) unified pathological grade and clinical stage, immunohistochemistry was used to detected the expression of MTA1, MMP-9and VEGF-C. SPSS19.0was used to analyze the relationship between the three and clinicopathological features,the correlation of the expression of MTA1and MMP-9,VEGF-C protein in gastric adenocarcinoma.Results:The positive percentage of MTA1, MMP-9and VEGF-C protein in gastric adenocarcinoma was76.7%(92/120)ã€71.7%(86/120)ã€64.2%(77/120) respectively, which was significantly higher than that in normal gastric mucosa tissues33.3%ã€36.7%and30.0%(P<0.05). The expression of MTA1, MMP-9and VEGF-C was significantly higher in gastric carcinoma invasion serous membrane, lymph node metastasis, clinical Ⅲ~Ⅳ than that not invasion serous membrane, no lymph node metastasis and clinical â… ï½žâ…¡(P<0.05), but there was no difference in patient’s age, gender and differentiation grade (P>0.05). The expression of the MTA1had significantly positive correlations with that of the MMP-9(r=0.352, P=0.002) and VEGF-C (r=0.306, P=0.012) in gastric adenocarcinoma.Conclusion:1.The expression of MTA1, MMP-9and VEGF-C was higher in gastric adenocarcinoma than normal gastric mucosa tissues, the expression of MTA1protein had significantly positive correlations with that of MMP-9and VEGF-C in gastric carcinoma. MTA1,MMP-9and VEGF-C may play an important role in the invasion and metastasis of gastric carcinoma.2.MTA1may promote the infiltration, invasion and lymph node metastasis of gastric carcinoma through regulation the expression of MMP-9and VEGF-C.The combined detection of MTA1, MMP-9and VEGF-C expression has important value to judge the grade malignance of gastric adenocarcinoma and provide the basis for adjuvant therapy. |
