Purpose:To detect the expression of p63and IKKβ in squamous cell carcinoma of cervix paired with adjacent normal tissue,and to explore its significance.Methods:We measured the mRNA expression of TAp63andâ–³Np63in a series21cervical squamous cell cancer paired with adjacent normal tissue by quantitative real-time polymerase chain reaction. The expression of P63ã€ANp63and IKKβ protein was detected in a variety of36cases of cervical squamous cell cancer paired with adjacent normal tissue by immunohistochemistry.Results:1. The relative level of TAp63andâ–³Np63mRNA in the tissue of cervical squamous cell cancer was0.156±0.040and0.264±0.067, respectively, which were markedly higher than those in adjacent normal tissues was0.061±0.019and0.032±0.008, respectively,and there were significant differences between two groups (P<0.05).2. The ration of the relative level ofâ–³Np63mRNA and that of TAp63mRNA in the tissue of cervical squamous cell cancer and adjacent normal tissues was1.801±0.654and0.576±0.173, respectively,and there were significant differences between two groups (P<0.05).3. The expression rate of P63protein in the tissue of cervical squamous cell cancer and adjacent normal tissues was91.7%(33/36)and33.3%(12/36) respectively, there were significant differences between two groups (P<0.05). The expression rate of P63protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor sizeã€tumor invision and tumor differentiation(P>0.05).4. The expression rate ofâ–³Np63protein in the tissue of cervical squamous cell cancer and adjacent normal tissues was88.9%(32/36)and36.1%(13/36) respectively, there were significant differences between two groups (P<0.05). The expression rate ofâ–³Np63protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor sizeã€tumor invision and tumor differentiation (P>0.05).5. The expression rate of IKKβ protein in the tissue of cervical squamous cell cancer and para-cancer tissues was100%(36/36) and50.0%(18/36) respectively, there were significant differences between two groups (P<0.05). The expression rate of IKKβ protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor sizeã€tumor invision and tumor differentiation (P>0.05).6. The expression rate of P63protein was positively correlated with the expression of IKKβ protein in the tissue of cervical squamous carcinoma and para-cancer tissues(r=0.591,P<0.05).7. The expression rate of ANp63protein was positively correlated with the expression of IKKβ protein in the tissue of cervical squamous carcinoma and para-cancer tissues (r=0.492,P<0.05).Conclusion:1. The expression ofâ–³Np63mRNA was much higher than TAp63mRNA in the tissue of cervical cancer, while the former was much lower than the later in para-cancer tissues, so the unbalance between TAp63andâ–³Np63may be potential factor to promote cervical cancer. 2. The expression of P63andâ–³Np63protein were both markedly higher than that in para-cancers tissues,but they had no obvious correlation with the tumor sizeã€invision and differentiation,P63perhaps associated with the occurrence of cervical cancer.3. The expression of IKKβ protein was markedly higher than that in para-cancers tissues,but it had no obvious correlation with the tumor sizeã€invision and differentiation, it perhaps associated with the occurrence of cervical cancer.4. The expression of P63andâ–³Np63protein were both positively correlated with IKKβ in the tissue of cervical squamous carcinoma paired with adjacent normal tissue, P63and IKKβ maybe have synergistic effect to promote cervical cancer. |
