| Objective:To observe the effect of Catheter-Based Renal Sympathetic Denervation (RDN) on the blood pressure, renal tubular function, renal tubular and interstitial morphology in diet-induced hypertensive canine model.Methods:(1)Hypertension model was established by feeding beagle canine with high-fat diet, twenty-eight Beagles (10-12months old) were randomly divided into normal control group (normal diet, n=6) and model group (fed with high-fat diet, n=22).(2) After12weeks of feeding, the blood pressures of canine were stable. The model group was then divided into operation group (renal sympathetic denervation) and sham-operation group (renal angiography). Renal angiography was re-performed at the3rd monthã€the6th month after surgery.(3)The weight were measured by electronic scales while blood pressure and heart rate were measured by noninvasive sphygmomanometers.(4)Urine specimens were obtained by bladder puncture at baseline (pre-modelling), pre-operative (12weeks after modelling) and post-operative (3rd months6thmonth), Using immunonephelometry measure urinary albumin, urinary αl microglobulin (urine al-MG), retinol binding protein in urine (urinary RBP), urinary N-acetyl-β-D-glucosidase (urinary NAG).(5)6months after operation, the animals were killed.Using high-performance liquid chemical method for the determination of local circulation and renal NE levels; using ELISA assay kidney Angâ…¡, using immunohistochemistry and western-blot to determination TGF-β1, a-SMA, E-cadherin, and using hematoxylin-eosin, Masson staining and electron microscopy to observe morphological changes in renal and renal interstitial.Results:(1) After12weeks of modeling, the SBP, DBP, MAP and body weight of the model group (high-fat group), were significantly increased,there exist significant difference when compared with the baseline and the control group(P<0.05);1,2,4,6months after RDN, the SBP, DBP and MAP of the surgical group were significantly lower than preoperative time, but also significantly lower than the sham group, the SBP, DBP, MAP of the sham group was significantly higher, the difference were statistically significant (P<0.05);(2) the levels of urinary albumin, α1-MG, NAG, RBP of the model group (high-fat group)were increased significantly, there exist significant difference when compared with the baseline and the control group(P<0.05);(3) In the high-fat group, Angâ…¡, TGF-β1, α-SMA were increased significantly, while E-cadherin was significantly lower, and with the high-fat group statistically significant difference compared (P<0.05), high-fat group radio group Angâ…¡, TGF-β1,α-SMA significantly reduced compared with sham group, E-cadherin increases, and the difference was statistically significant (P<0.05);(4) fibrosis exist in the high-fat group,and sham surgery group was significantly than surgery group. Conclusions:(1) RDN can significantly lower SBP, DBP, and MAP in diet-induced canine hypertension.(2) RDN is safe.It will not damage the kidney.It can delay the renal tubules and interstitial fibrosis and improve kidney function.Figure18, Table18,Reference93... |
