ObjectiveThrough the study of serum calcium, phosphorus, osteocalcin, urinarydeoxypyridinoline, urine creatinine, carboxyterminal telopeptide of type Icollagen, Type I collagen cross-linked N-terminal peptide, and bone tissuepathological changes of bone mineral density af Erxian Decoction on castratedrats, and discussion of Erxian Decoction on osteoporosis in castrated the roleand the mechanism of rats, increase the theoretical basis for clinical therapy.Methods503-month-old female SD rats were randomly divided into five groups,were sham operation group, model group, estrogen group, Erxian decoctionhigh dose group, Erxian decoction low-dose group. In addition to the shamgroup, are to be ovariectomy.5days after administration, the sham group andmodel group were given normal saline, low dose group was given1.79mlmedicine (including3.04g crude drug), herbal medicine high-dose group wasgiven1.79ml (including12.16g crude drug), once a day for12weeks. The firstsix weeks of fasting to take measurements of urinary calcium (U-Ca),deoxypyridinoline (DPD), urine creatinine (U-Cr), U-DPD/Cr; first12weeks ofserum calcium (S-Ca radioimmunoassay), serum phosphorus (SP),Osteocalcin, carboxyterminal telopeptide of type I collagen (PICP), Type Icollagen cross-linked N-terminal peptide(NTX); dual-energy X-ray absorptiometry diagnostic testing left femur bone density. Femur pathologicalbiopsy.ResultsEffects of calcium, phosphorus, serum osteocalcin content:Calcium content:compared with the model group, sham operation group, estrogen group, ErxianDecoction group were increased (P<0.05). Compared with the ER XianDecoction high dose group, no significant differences in estrogen group (P>0.05). Decreased content Erxian Decoction low dose group and estrogen group(P<0.05). There was no significant difference (P>0.05) S-P. Osteocalcin content:compared with the model group, sham operation group, estrogen group, ErxianDecoction group were significantly reduced (P<0.01). Compared with the ERXian Decoction high dose group, estrogen group and ER Xian Decoction lowdose group content were increased (P<0.05).Deoxypyridinoline, creatinine, urinary deoxypyridinoline/creatinine,carboxyterminal telopeptide of type I collagen, Type I collagen cross-linkedN-terminal peptide: compared with the model group, sham operation group,estrogen group, Erxian Decoction deoxypyridinoline, creatinine, urinarydeoxypyridinoline/creatinine, Type I collagen cross-linked N-terminal peptidedecreased significantly (P<0.01); carboxyterminal telopeptide of type Icollagen was significantly increased (P<0.01). Compared with the ER XianDecoction high dose group, estrogen group deoxypyridinoline, creatinine,urinary deoxypyridinoline/creatinine had no significant differences (P>0.05),carboxyterminal telopeptide of type I collagen increased (P<0.05), Type Icollagen cross-linked N-terminal peptide decreased significantly (P<0.01); twofairy Decoction low dose group deoxypyridinoline, creatinine, urinarydeoxypyridinoline/creatinine, Type I collagen cross-linked N-terminal peptideincreased significantly (P<0.01), carboxyterminal telopeptide of type I collagen was significantly decreased (P<0.01).Effect of bone density: compared with the sham operation group (groupSham), model group (group OVX) bone mineral density decreased significantly(P<0.01), suggesting that the animal model of osteoporosis in rats. Comparedwith the model group, estrogen group, Erxian Decoction group, the bone densitywas significantly increased (P<0.01). Compared with the ER Xian Decoctionhigh dose group, estrogen group and ER Xian Decoction low dose group ofbone density was significantly decreased (P<0.01). Show Er Xian decoctiontreatment has obvious dose-dependent.Biopsy: model group: trabecular bone thinning, slender, the gap increases,fewer connections between trabecular bone, arranged clutter. Estrogen andMedicines display trabecular bone compared with the model group stout,shorten the gap between the trabecular bone, form a complete, compactstructure, bone plates, arranged in order.Pathological section: sham operation group (group Sham) visible bonetissue structural integrity, trabecular bone density stout, number, shape rules,the gap is moderate, neat rows; shown in Figure2, the model group (groupVOX), trabecular bone thin, sparse, inter trabecular fracture and separation, gapincreased, arranged in disorder, bone lamellar structure is loose, osteoporosis isobvious; as shown in Figure3, estrogen group (group EST), trabecular stout,complete structure, regular shape, compact and trabecular bone, trabecularnumber the Union more than model group; shown in Figure5, Erxian soup highdose group (EXT/L group) can be bone trabecula dense abundance, spacing isnarrow, morphological structure of regular, width and gap and sham operationgroup and estrogen group approachesConclusionsErxian Decoction to castrated rats serum calcium, Osteocalcin, carboxyterminal telopeptide of type I collagen, increased bone mineral density;osteocalcin, urinary deoxypyridinoline, urinary creatinine reduction; urinarydeoxypyridinoline/creatinine decreased; Type I collagen cross-linked N-terminalpeptide decreased histopathological improvement. Erxian Decoction canpromote bone formation, inhibit bone resorption, maintain bone remodeling ofbone metabolism, increase bone density, so as to effect and protective effect ofanti osteoporosis bone.
