ObjectTo analyze the clinical data between EOS-NP and non-EOS-NP, detect theexpression of IL-25, TSLP, IL-17A in EOS-NP, non-EOS-NP, the nasal mucosaof inferior turbinate and analyze their correlation, in order to investigate the roleof IL-25, TSLP, IL-17A in the pathogenesis of EOS-NP and non-EOS-NP.MethodTo divide the patients of nasal polyps into EOS-NP group and non-EOS-NPgroup according to the result of HE staining, analyze the Symptom scores,nasal endoscopic score, sinus CT score, SPT result of the two groups. Westernblot was used to detect the protein expression of IL-25, TSLP, IL-17A in NPtissues, RT-PCR was used to detect mRNA expression of IL-25, TSLP, IL-17Ain NP tissues, ELISA was used to detect expression of TSLP, IL-25, IL-17A inperipheral blood of NP patients, and analyze the correlativity among IL-25,TSLP and IL-17A.Result1.69cases of patients with NP were divided in to two groups by HEstaining, we find33cases are divided into EOS-NP group, account for47.8percent of the NP patients, and36cases are divided into non-EOS-NP group,account for52.2percent of the NP patients.2. EOS-NP group has the higher nasal endoscopic grading, sinus CTgrade, SPT positive rate than non-EOS-NP group, a statistically significant difference(P<0.05), the difference in age, sex, and the length of disease between the twogroups have no statistics significance (P>0.05).3. The protein and mRNA expression of IL-25, TSLP and IL-17A in NPpatients are higher than the control group, with difference statistical significance(P<0.05), the protein and mRNA expression of TSLP and IL-25in EOS-NPgroup is higher than non-EOS-NP group, a statistically significant difference(P<0.05), the protein and mRNA expression of IL-17A in EOS-NP group ishigher than non-EOS-NP group, a statistically significant difference (P<0.05).The IL-25is positive correlated with TSLP of protein and mRNA in EOS-NPgroup and non-EOS-NP group, the TSLP is negative correlated with IL-17A ofprotein and mRNA in EOS-NP group and non-EOS-NP group, and the IL-25isnegative correlated with IL-17A of protein and mRNA in EOS-NP group andnon-EOS-NP group.4. The expression of IL-25and TSLP in peripheral blood of NP is higherthan the control group, with difference statistical significance(P<0.05), theexpression of IL-25and TSLP in peripheral blood of EOS-NP group is higherthan non-EOS-NP group, with difference statistical significance(P<0.05). TheIL-25is positive correlated with TSLP in peripheral blood of EOS-NP andnon-EOS-NP group, there is no correlation between TSLP and IL-17A, there isno correlation between IL-25and IL-17A in two groups.5. The protein expression of IL-25in NP tissues is positive correlated withthe expression of IL-25in peripheral blood of NP patients (P<0.05), the proteinexpression of TSLP in NP tissues is positive correlated with the expression ofTSLP in peripheral blood of NP patients (P<0.05), there is no correlationbetween the protein expression of IL-17A in NP tissues and the expression ofIL-17A in peripheral blood of NP patients (P>0.05). Conclusion1. Compared with non-EOS-NP, EOS-NP has the higher pathologicalchanges degree and range and the higher SPT positive rate.2. IL-25and TSLP play important roles in systemic immunity of EOS-NPand non-EOS-NP, the pathogenesis of EOS-NP is closely related to IL-25andTSLP.3. IL-17A play important roles in local mucosa immunity of EOS-NP andnon-EOS-NP, the pathogenesis of non-EOS-NP is closely related to IL-17A.
