Research Of The Regulation Of Saikosaponin D On GRβmRNA Of Glucocorticoid Resistance IdiopathicThrombocytopenic Purpura

Objictive:to obsrrve the expressions of glucocorticoid receptor-β in peripheral blood mononuclear cells(PBMC) and glucocorticoid resistance in patients with idiopathic thrombocytopenia purpura,and its intervention by saikosaponin-d(SSd)。Method:Real-time PCR was used to detect of GRβmRNA in PBMC from30patients with ITP(glucocorticoid-sensitive,n=18; glucocorticoid-resistant,n=12),12glucocorticoid-resistant patients treated with SSd and20healthy people (control group)。Result:There were no significant differences in PBMC GRβmRNA levels in between the glucocorticoid-resistant and the control groups。The expression of GRPmRNA in the resistant group of ITP was significantly higher than that in both sensitive group and control group,P<0.05。The expression of GRβmRNA in the resistant group of ITP treated with SSd was significantly lower than that in resistant group, P<0.05。Conclusion:The up-regulated expression of GRβmRNA may associated with glucocorticoid resistance in patients with ITP。SSd can decrease the up-regulated expression of GRβmRNA in glucocorticoid resistance patients with ITP o...
