| Part1Research Background[Abstract] Malignant melanoma is a malignant tumor that originated from neural crest melanocytes. Surgery, chemotherapy, radiotherapy, gene therapy and immunotherapy were always used in the treatment of malignant melanoma. But the curative effect was not ideal. Traditional Chinese medicine (TCM) and Chinese herbal medicine are characteristic treatment method and precious resources of our country. Some herbal medicines played an important role in skin whitening through inhibiting tyrosinase activity in melanocytes. And they have already been used in the treatment of skin diseases and cosmetics researches. The malignant melanoma cells always accompany with the increase of tyrosinase activity. Therefore, the anti-melanoma effect of these whitening Chinese herbal medicines is worthy of studying. Part2Primary Study on Effect of Five Chinese Herbal Medicine Monomers on Inhibiting Proliferation and Tyrosinase Activity of Human Malignant Melanoma A375Cells in Vitro[Abstract] Objective:To study the effect of five Chinese herbal medicine monomers on inhibiting proliferation and tyrosinase activity of human malignant melanoma A375cells.Methods:Five kinds of Chinese herbal medicine monomers (Arbutin, Danshensu, Aloe-emodin, Curcumin and Tanshinone IIA) were selected. MTT colorimetric method was used to measure the inhibiting effect on proliferation of these Chinese horbal medicine monomers in human malignant melanoma A375cells in vitro, while the rate of oxidation L-DOPA was used to detect the inhibition of tyrosinase activity. Results:All of the5traditional Chinese medicine monomers showed dose-dependent down-regulation of A375cells’proliferation at certain concentration range (P<0.05). Arbutin, Danshensu, Aloe-emodin and Curcumin could inhibite tyrosinase activity. However, after48hr, high concentration of Arbutin and Danshensu still inhibited the tyrosinase activity with significant statistic difference(P<0.05), while no significant statistic difference had been observed in high concentration of Aloe-emodin and Curcumin. Tanshinone IIA hadn’t showed obvious effect of inhibiting tyrosinase activity. Conclusion:There is a concentration-dependent relationship between these5kinds of Chinese herbal medicine monomers (Arbutin, Danshensu, Aloe-emodin, Curcumin and Tanshinone IIA) and the inhibition effect of A375cells proliferation. Also, certain concentration of Arbutin and Danshensu could inhibit tyrosinase activity significantly, while Aloe-emodin, Curcumin and Tanshinone IIA hadn’t showed obvious inhibition effect of tyrosinase activity. We speculated that the inhibition effect of tyrosinase activity could be a conceivable mechanism for the anti-melanoma function of Arbutin and Danshensu. Aloe-emodin, Curcumin and Tanshinone IIA could inhibit A375cells proliferation though other mechanisms. |
