Objective This topic will be based on osteoclasts in vitro co-culture with bone plates, titanium particles with different concentrations and different concentrations of icariin intervention explore icariin in the process of osteoclast activity induced by titanium particles intervention mechanism and tried to dig single herb for the prevention and feasibility of aseptic loosening of prosthetic treatment.Methods The experiment chosen to RANCAL and M-CSF as an inducer of macrophage (RAW264.7) induced stimulation of osteoclasts co-cultured with bone plates, artificial joints wear after adding titanium particles inactivated saline dubbed microparticle suspension was added osteoclasts, bone cell activity was observed under the microscope broken then added at different concentrations (10-5mol/L,5×10-5mol/L,10-6ml/L,10-7mol/L) after comparison with the former icariin. After statistical processing, analysis icariin intervention mechanisms titanium particles induced osteoclast activity areas.Results After the microparticles to stimulate bone resorption lacuna board and osteoclasts than without any treatment group was significantly increased, the image size is large lacunae analysis (P<0.05). Icariin intervention group resorption area less than the control group and the model number of osteoclasts (P<0.05), with10-5mol/L concentration group the most significant, followed by5×10-5mol/L,10-6ml/L. Among them, the model group and the10-7mol/L group had no significant difference.Conclusion Titanium wear particles can induce bone resorption and promoting osteoclast activity. This icariin can inhibit osteoclast activity induced by titanium particles, thereby suppressing periprosthetic osteolysis, with a more important role in the prevention and treatment of aseptic loosening of artificial joints.
