Objective: To Investigate The effect induced by LY294002alone and combined withNedaplatin(NDP) on human cervical carcinoma HeLa cell LY294002sensitizationeffect of NDP,and its possible mechanism in vitro。Methods:(1)Human cervical carcinoma HeLa cell were treated with LY294002,Nedaplatin(NDP) or LY294002combined with Nedaplatin(NDP) in vitro.The cellgrowth was assessed by MTT assay.(2)The expressions of intracellular PI3k, p-PI3k,Akt, p-Akt, Bcl-2, and Caspase-3proteins in human cervical carcinoma HeLa cellwere analyzed by western blotting.Results:1.LY294002,NDP can inhibit the growth of cervical cancer HeLa,with theincrease of drug concentration, cell inhibition rate increased significantly, P <0.05,the difference is statistically significant.2.LY294002could enhance the synergism of HeLa cells to NDP. LY294002, NDPsingle-agent applications of cell inhibition rate significantly lower LY294002andNDP combination inhibition rate, P <0.05, the difference is statistically significant.Use the formula to calculate joint index, tip combination, has a synergistic effect.3.Western Blot analysis results showed:LY294002and NDP can decrease theexpression of Bcl-2protein and raising Caspase-3protein expression.Compared withthe LY294002or NDP alone treatment group, the combination treatment group’sBcl-2protein expression were decreased but the expression Caspase-3proteinwere increased.4.PI3K, Akt proteins were expressed in human cervical carcinoma HeLa cells,LY294002could inhibit the expression ofp-PI3K,p-Akt proteins. Conclusions:(1)The proliferation of PI3K inhibitor LY294002and NDP wasinhibited by human cervical carcinoma HeLa cells, and the effect was dose-dependentand time-dependent.(2)Followed by the increase of the concentration of LY294002,LY294002couldenhance the synergism action of LY294002to NDP,enhance apoptosis of cells andincrease the sensibility of HeLa cells to NDP.(3)LY294002inhibited proliferation and induce apoptosis, which may be related toreduced expression of Bcl-2protein and Increased Caspase-3protein expression.(4)LY294002inhibited the expression of p-PI3K and p-Akt.The results of thisexperiment demonstrated that the suppression of PI3K/Akt signal transductionpathway is perhaps the most important mechanism in the treatment of cervicalcarcinoma with LY294002.
