| Objective The aim of this study is to assay the anti-tumor effect ofTerpinen-4-ol in cell line A549, and also to explore its possible mechanismrelated pathway.Methods A549cultures were incubated with various concentrations ofTerpinen-4-ol,The MTT assay was used to observe the inhibitory effect ofTerpinen-4-ol on the growth of adenocarcinoma cell A549and its IC50. Cellgrow ability was determined by CCK8colorimetry. The ultrastructure ofA549cells were observed by transmission electron microscopy before andafter Terpinen-4-ol treatment. The changes of cell cycle, apoptosis andintracellular free calcium concentration were measured by flow cytometry.Real-time fluorescent quantitative PCR and Western blot were performed todetect BECN1,LC3,BCL-2and Caspase3expression changes.Results After Terpinen-4-ol treatment for24h,MTT assay showed thatthe IC50value of A549cells was0.067%v/v. The growth curves oftreatment groups were significantly smooth than the control group. Theformation of autophagosome increased after treatment with Terpinen-4-ol.The results of flow cytometry showed that the cell cycle was arrested in Sphase,Terpinen-4-ol could induce apoptosis of A549cell, The intracellular calcium concentrations in treatment groups were significantly higher thanthe control group(P<0.05). Low dose group and high dose group restrainedthe growth of the transplantation tumor obviously (tumor inhibitory rate53.33%and77.76%). Realtime-PCR and Western blot analysis showed thatTerpinen-4-ol inhibited the mRNA and protein levels of BECN1, LC3,Caspase3, whereas reduced the expression of BCL-2(P<0.05).Conclusion Terpinen-4-ol had inhibitory effect on the proliferation ofA549cells in vitro and in vivo. |
