| Objective:The blood transfusion remains to be an irreplaceable method for the therapy of many diseases, as well as the direct way for patients to obtain blood. Yet some problems still exist in the safety of blood transfusion. In order to guarantee the safety of blood transfusion and avoid diseases spreading via blood, we must intensify the blood screening efforts. Therefore, it is extremely important to add new biomedical markers for screening tests. The Neopterin(NPT) concentrations can indirectly reflect the immune function of macrophages in body, thereby indirectly reflect the potential possibility of the body infected by the virus. The NPT concentions in serum changes earlier than other screening markers, therefore it has become a hot research topic in blood screening. This research is meant to establish a normal reference range of serum NPT among local healthy populatio, and compared with other blood screening index, discuss the possibility and significance about whether serum NPT concentrations can be an index of screening blood donor, by detecting the serum NPT concentrations of blood donors.Methods:The serum NPT concentrations of12562random blood donors were detected by ELISA to obtain the normal reference range of NPT in healthy population in Yue Yang, and the data derived from the routine tests of blood donors were compared.The routine blood screening including ALT, HBsAg, anti-HCVã€anti-HIV and anti-TP test, velocity method was used to detect ALT of the blood donors for further test, and Enzyme linked immuno-sorbent assay (ELISA) method was used to detect Hepatitis B virus surface antigen (HBsAg), Hepatitis C virus antibody (anti-HCV), HIV antibody (anti-HIV), and Treponema pallidum antibody (anti-TP). Analyzing the relationship between the causes of different results and the concentration of serum NPT.Results:1. By selecting2000healthy people, calculate the normal reference range of the serum NPT concentration in the healthy populations of the region is less than7.95nmol/L2. Among12562random blood donors, unqualified rate of neopterin concentration screening was3.02%(380/12562), which is higher than unqualified rate1.75%(220/12562) detected by normal blood screening, The unqualified rate between routine blood screening and serum neopterin concentration screening is of significant difference(P<0.01).3. The unqualified rates of the serum NPT concentration has no significant difference between different sex, age, occupation, literacy or BMI of people, NPT can be used as one of the specific index for early diagnosis of infectious diseases.4. There are220blood donors fail in routine screening out of380unqualified donors, and other160blood donors are also unqualified in serum NPT concentration screening though they are qualified in routine screening. Through tracking and follow-up visit, it can be confirmed that 8out of160blood donors once infectioned the pathogens or have potential dangers.Conclusion:1. The normal reference range of serum neopterin concentrations of eligible blood donors in yueyang is less than7.95nmol/L (the normal value of99.5%, unilateral)2. The serum concentration of the unqualified blood donors in routine screening were all increased.3. The concentration of serum NPT is independent of nonspecific factors such as gender, age, occupation, literacy, BMI and etc, Can be used as one of the specific index for early diagnosis of the infection diseases.4. Serum neopterin concentration detection can be used in screening of blood donors with potential danger, In order to further improve the safety of blood transfusion. |
