We could explore the new mechanism of Valsartan in pressing down the blood and to observe the protective effect of ACE2in the target organ(kidney and brain) damage caused by hypertension.Methods:Twenty-four12week-old male SHR were randomly divided to SHR group(n=12)and valsartan group(n=12). Twelve12week-old male WKY rats were served as control group. Valsartan group was given daily dose of30mg/kg valsartan for10weeks and the other two groups were given equal saline. After10weeks, kill all the rats, take out the kidney and brain. Carry out the following index test through the following experimental methods:1ã€To observe the changs of organizational structure in the kidney and brain of spontaneously hypertensive rats through H E staining method2ã€Detect the expression of ACE2protein in kidney and brain of spontaneously hypertensive rat by immunohistochemistry.3ã€Detect the expression of mRNA in kidney and brain of spontaneously hypertensive rat by RT-PCR.Results:1ã€The systolic blood pressure(SBP) of SHR was significantly hihger than the control group in the first detection. After10weeks detect the SBP again, The SBP of control group the SBP without apparent change.The SBP of valsartan group was markedly decreased,but higher than the control group.And it was obviously increased in SHR group.2ã€It is nomal about organizational structure in the kidney and brain of spontaneously hypertensive rats in the control group.In the SHR group,there are obviously pathological change. And the change of valsartan group fall in the between.3ã€The resault of immunohistochemistry indicated the ACE2protein expressed in the kidney and brain cell.The ACE2protein expression of SHR group and valsartan group were lower than the control group, but the valsartan group was higher than the SHR group. 4ã€The resault of RT-PCR indicated the ACE2mRNA expression in the kidney and brain of SHR group was decreased compared with the control group, the valsartan group was increased.Conclusions:1ã€The blood pressure of the SHR was significantly hihger than the WKY rats, Valsartan was able to significantly decreased the SBP.2ã€The ACE2mRNA and ACE2protein expression of SHR were markedly lower than WKY rats.
