ObjectiveAutologous fat transplantation has been widely used in plastic and reconstructivesurgery in recent30years, It has become an important method of soft tissue filling.But the simple autologous fat transplantation has high absorption rate and lowsurvival rate, thus limiting its further promote using. This experiment used adiposestem cells stem cells mixing with sodium hyaluronate, type I collagen in theappropriate proportion in order to build stem cell-scaffold complexes,then injectedinto nude mice subcutaneous soft tissue and explore its effects on subcutaneous softtissue vascularization, and provide a basis for screening adipose stem celltransplantation and related biological scaffold to provide a theoretical basis forclinical application.MethodsADSCs’separation and inoculationCollagenase digestion method is used to extract the adipose derived stem cells, and isolate the primary ADSCs at4×105cells/m concentration to passage in DMEM,collect the three cells as seed cells. The third generation cells were seeded in culturemedium and added to the general inducer fully adherent (106mmol/L dexamethasone,10mg/L insulin,0.5mmol/L3-isobutyl-1-methylxanthine and0.2mmoL/Lindomethacin) for2weeks.This experiment of the five groupsGroup A(ADSCs combinding with fat particles), Group B (ADSCs combindingwith diluted Hyaluronic Acid), Group C(ADSCs combinding with type Icollagen), Group D(pure ADSCs), Group E(pure fat particles). Each nude musewas marked5points. Five groups of reagents were injected into the subcutaneousstratum nude mice. The nude mice were harvested after3months, the implants werecompletely removed and the correlation detection and analysis were conducted aboutit.ResultsGeneral observationWhen drawn, we can see a group of tissue blocks in injected subcutaneously of nudemice, clear boundaries, encapsulated, easily separated.The sizes and shapes ofadipose tissue were irregular.microvessel densityGroup A,group B and group C was higher than group D and group E, and hadsignificant statistically differences (P<0.01), there were no significant statisticallydifferences between group D and group E(P>0.05), there were no significantstatistically differences among group A, group B and group C (P>0.05).Comparison of survival (%)Group A,group B,group C was higher than group D and group E, and had significantstatistically differences (P<0.01), group D was higher than group E, and hadsignificant statistically differences (P<0.01), there were no significant statisticallydifferences among group A, group B and group C (P>0.05).Determination of wet weight Group A,group B and group C was higher than group D,and had significantstatistically differences compared with group D(P<0.01), there were no statisticallysignificant differences between group D and group E, there were no statisticallysignificant differences among group A,group B and group C (P>0.05).Conclusions1. ADSCs have low immunogenicity, does not have a pyrogenic reactions andinflammation via local injection.2. ADSCs having pro-vascularization.3. Adipose tissue, hyaluronic acid, collagen can accelerate the vascularization ofADSCs, and the three scaffolds are no significant differences.4. ADSCs and biological scaffold has a synergistic effect.5. ADSCs compbinding with related cellular scaffold are feasible to be regarded as ainjectable material, hope to provide a new injectable cosmetic procedure for theplastic cosmetic surgery.
