Study On Purification And The Effects On Hyperlipidemia Model Rats Of Polyphenols From Sabina Vulgaris Antoine

Sabina vulgaris is a kind of evergreen perennial creeping shrub,which spreads mainly in Tianshan Mountains,Altai Mountains,and Junggar Basin of Xinjiang.In Uygur Medicine,the twigs and fruits of Sabina vulgaris is used as a medicinal herb.There are several specieces of chemical compositions including volatile oil, flavonoids, Lignans and coumarins,and so forth.In this research,we applied homogenate extraction, in which acetone was used as solvent, to extract the polyphenols of Sabina vulgaris, and then concentrated to get the extract.Folin-Ciocalteu method was adopted to determine the content of Sabina vulgaris polyphenols.The condition of Folin-Ciocalteu method was optimized.Then the standard curve was drawed,in which gallic acid was used as reference substanceIn this research, macroporous resin was used for the purification of Sabina vulgaris polyphenols.From9kinds of macroporous resin, HPD-700was selected as the best macroporous resin for this purification,and the purification condition was also optimized.After purification,the purity of Sabina vulgaris polyphenols increased from0.53%to9.95%.The research of the antioxidant activity in vitro of Sabina vulgaris polyphenols indicated that Sabina vulgaris polyphenols have relatively strong Scavenging capacity. The inhibition ratio of DPPH free radical and hydroxy radical is62.93%and26.29%,the inhibition capacity of superoxide anion is151.83U/mL,the reducing capacity for Fe3+is30.09VCEAC(μg/mL),the total antioxidant capacity was72.68U/mL.Hyperlipoidemia rat model was induced with high fat diet,and used for evaluating the influence of Sabina vulgaris polyphenols.The results indicated that compared with the model group,Sabina vulgaris polyphenols could decrease the weight of hyperlipoidemia rat model.Further more,the TG,TC and LDL-C in hyperlipoidemia model rat blood was declined,too.The HDL-C increased slightly.The experimental result indicated that Sabina vulgaris polyphenols showed antioxidant activity and the effect of lipid-lowering.
