| Objective:To investigate the association between the peroxisome proliferator-activated receptor y2Pro12Ala gene polymorphism with type2diabetes or diabetic nephropathy,and analysis the risk factor for diabetic nephropathy.Methods:According to WHO1999criteria,248type2diabetic inpatients were selected(male134cases,female114cases,including diabetic nephropathy)from the medical ward of Endocrine during December2011to December2013.The control group was composed of healthy check-up outpatients(male65cases,female60cases),and the clinical data were collected.Genomic DNA was extracted from peripheral blood leucocytes of the participants. We screen the Pro12A1a polymorphism by means of PCR-RFLP in all subjects and identification the mutation by directly sequenced the PCR production. At last we compaired the difference of frequency in allele and genotype among groups.Results:1.The frequency of the Pro12Ala mutation was significantly lower in diabetic subjects than in non-diabetic subjects(P<0.05).2.In the diabetes group,the LDL-C and HDL-C have significantly difference among different genotypes(P<0.05).3.The frequency of the mutation was significantly lower in diabetic nephropathy group than in non-diabetic nephropathy group(P<0.05).4.The frequency of the Pro12Ala mutation did not differ significantly among the obese group (P>0.05)Conclusion:1. The peroxisome proliferator-activated receptor y2Pro12Ala gene polymorphism may be associated with type2diabetes or diabetic nephropathy.2. Allele A is the protective factor of type2diabetes or diabetic nephropathy.3. The peroxisome proliferator-activated receptor y2Pro12Ala gene polymorphism have effects on lipid metabolism in type2diabetes mellitus, but have no association with obesity.4. Total cholesterolã€creatinine and systolic blood pressure are independent risk factors for diabetic nephropathy. |
