Objective(1)Jinhuang Powder-Polyurethane dressing is based on the famous prescription "Ruyijinhuangsan" of traditional chinese orthopedics and used medical polyurethane as drug carrier material. As a suitable material for temporary wound coverage, it’s effect on anti-inflammatory, relieving pain, promoting granulation, adsorbing wound secretion and keeping the wound relative moist.(2)The preliminary test of the physical, drug loading performance of Jinhuang Powder-Polyurethane dressing.(3) Observe the anti-inflammatory, eliminating necrotic tissues and skin irritation of Jinhuang Powder-Polyurethane dressing used to acute wound of guinea pigs.Methods(1)Weighing the drugs of Jinhuang Powder medicine accurately according to the proportion of the Chinese Pharmacopoeia and taking10times the95%ethanol as solvent to extract the drug composition. Using immersion method to make the Jinhuang Powder-Polyurethane dressing.(2)The following tests have been taken of the dressing:morphological observation, adsorption test, liquid preservation and drug loading performance.(3)Take30ordinary Hartley guinea pigs to establish the experimental model of acute soft tissue contaminated wound, and respectively covered by Jinhuang Powder-Polyurethane dressing, polyurethane materials and ordinary yarn block. After three days check the:①Observing morphology of wounds, pus moss, amount of blood stasis;②Collecting and culturing the wound bacterium, observing colony morphology, and counting the staphylococcus colony;③Using microscopic to observe the histological changes of wound tissue;④Take any five horizons in light of microscope at400times, and counte fibroblasts, macrophages, neutrophils number and compare the average.(4)Taking8guinea pigs to design back scratching wounds, respectively applied by Jinhuang Powder-Polyurethane dressing and ordinary gauze, to observe the skin irritation in72h.Results(1) Development of the polyurethane material soaked in medicine is higher than the one without medicine in Micro-CT, and the medicine particulate at all levels is evenly cloud-like distributed.(2)At the beginning1st min of the soaking, polyurethane sucting quickly and reach the70%of its max-amount, then gradually leveled off, almost saturated after5mins of soaking, and the stable suction reach15~20times of its own weight. There is no significant difference in the absorption of water, saline and blood.(3)HPLC shows that the content of curcumin in dry Jinhuang Powder-Polyurethane dressing is more than90%in Jinhuang Powder stoste, which prove polyurethane material has good drug loading capability.(4) Three days later, the wound in Jinhuang Powder-Polyurethane dressing group is dry and clean, ruddy in color, little bleeding, and has more fibroblasts and macrophages, less neutrophils, which is better than the other groups.(5)Skin irritation test shows that the Jinhuang Powder-Polyurethane dressing used on the skin has no irritation.ConclusionPolyurethane can absorb higher than nearly15-20times its own weight of liquid, having good suction performance. Using immersion method to make the Jinhuang Powder-Polyurethane dressing can have high drug-loading capacity, which applied to the acute wound of guinea pig can play efficacy of anti-inflammatory and eliminating necrotic tissues, without irritation to the skin.
