Objective:To investigate the expression and significance of MCM7、Skp2、P27、Rb proteins in normal cervical squamous cells, cervical intraepithelial neoplasia(CIN) and cervical squamous cell carcinoma(CSCC). Exploring the possible mechanism of them in CIN and CSCC。Method:Experimental specimens were collected by the department of pathology in Tianjin Medical University General Hospital from January2009to March2013.Then selected10cases of normal cervieal squamous epithelium,90cases of CIN(including30cases of CIN Ⅰ,30cases of CINⅡ,30cases of CINⅢ),30cases of CSCC. The expression of MCM7、Skp2、P27、Rb were examined by immunohistochemical technique(PV6000). The data of this study was analyzed by SPSS19.0software package(Kruskal-WallisH test、Mann-WhitneyU test and Spearman correlation analysis)Results:1. The morphologic characteristic stained of MCM7、Skp2、P27、Rb:These four proteins positive staining were located in nucleus.2. The expression of MCM7protein gradually increased from the normal to CIN to CSCC, there was significant difference in each group (H=66.751, P<0.001). The expression of CIN Ⅰ was no significant difference with the normal group (P>0.01), the expression of CIN Ⅱ,CINⅢ, CSCC group were higher than the normal group(P<0.01). The expression of CIN Ⅱ was higher than the CIN Ⅰ group (P<0.01).The expression of CSCC was higher than all CIN group(P<0.01).3. The expression of Skp2protein gradually increased from the normal to CIN to CSCC, there was significant difference in each group (H=43.492, P<0.001) There was no significant difference between the normal group and the CIN I group (P>0.01),the expression of CIN Ⅱ,CINⅢ, CSCC group were higher than the normal group(P<0.01).There was no significant difference between the CINⅠ andCⅡ group, CINⅡ and CINⅢ group(P>0.01).The expression of CSCC was higher than all CIN group (P<0.01).4. The expression of P27protein gradually reduced from the normal to CIN to CSCC, there was significant difference in each group (H=48.945,P<0.001).The expression of CIN Ⅰ was no significant difference with the normal group (P>0.01), the expression of CIN Ⅱ, CINⅢ, CSCC group were lower than the normal group (P<0.01). The expression of CIN Ⅱ was lower than the CIN Ⅰ group (P<0.01),the expression of CINⅢ was lower than the CIN Ⅱ group(P<0.01).There was significant difference between the CSCC and CIN Ⅰ, CIN Ⅱ group(P<0.01).5. The expression of Rb protein gradually reduced from the normal to CIN to CSCC, there was significant difference in each group (H=54.140,P<0.001).There was significant difference between the normal group and the CIN Ⅱ,CINⅢ, CSCC group (P<0.01).The expression of CIN Ⅱ was lower than the CIN Ⅰ group(P<0.01),the expression of CINⅢ was lower than the CIN Ⅱ group(P<0.01).There was significant difference between the CSCC and CIN Ⅰ, CIN Ⅱ group(P<0.01),but there was no significant difference between the CSCC and CIN3group(P>0.01).6. The expression level of MCM7was negatively correlated with P27and Rb (r=-0.498, p<0.001; r=-0.427,p<0.001); The expression level of Skp2was negatively correlated with P27and Rb(r=-0.540, p<0.001; r=-0.491, p<0.001);The expression level of P27was positively correlated with Rb(r=0.930, p<0.001).Conclusion:1. The expression degree of MCM7and Skp2were increased gradually from CIN to CSCC, they probably play some important role in the process of CIN to CSCC. Detection of MCM7may be helpful to distinguish between low grade CIN and high grade CIN lesions.2. The expression degree of Rb and P27were reduced gradually from CIN to CSCC, The declining and abnormal expression of Rb and P27probably play some important role in the process of CIN and CSCC. The expression level of Rb was positively correlated with P27, they may cooperate in process of the development and progression of cervical cancer.3. The expression level of MCM7was negatively correlated with P27and Rb, the reducing or missing expression of P27and Rb may be up-regulate the expression level of MCM7by some ways. They can promote the development of CIN and CSCC.4. The expression level of Skp2was negatively correlated with P27and Rb, Skp2may be a negatively regulating factors of P27and Rb, it might participate the malignant transformation of cervical squamous cells carcinoma by the ubiquitin proteasome path-way(UPP).5. There was a significant difference for the expression of MCM7、Skp2、P27、Rb in each group of CIN and cervical squamous cell carcinoma (P<0.001).To detect the MCM7、Skp2、P27、Rb separately or jointly may be helpful to the auxiliary diagnosis of CIN and CSCC, the application prospect of it needs further study.
