ObjectiveBy testing expressions of VEGF-C and Ki-67in breast infiltrative ductalcarcinoma and tissues beside tumor, through analysis of differences inexpressions and relationship between clinicopathological factors, roles ofVEGF-C and Ki-67in the occurrence,development and metastasis of breastinfiltrative ductal carcinoma are to be explored.MethodologyImmunohistochemistry SP was adopted to test VEGF-C and Ki-67proteinsof120breast infiltrative ductal carcinoma and tissues beside tumor. Statisticalanalysis was conducted with the data using Wilcoxon Kruskal-Wallis rank-sumtest and Spearman rank correlation analytical method.Results1.Expression of Ki-67protein, appearing as brown yellow granules islocated in the cell nucleus. Expression rates in breast infiltrative ductalcarcinoma and tissues beside tumor are55%and5%respectively, thedifferences of which are statistically significant(χ2=17.17, P<0.05).Highexpression rates of Ki-67protein in breast cancer histological grades I、Ⅱ、 IIIare14.29%、47.47%、78.38%respectively, the differences of which arestatistically significant (P<0.05) and positively correlated to histological gradeof tumors (r=0.35, P<0.05). Expression of Ki-67has not been found to beaffected by patients’ age, tumor size and lymph node metastasis.2. Expression of VEGF-C protein, appearing as brown granules is locatedin the endochylema of breast infiltrative ductal carcinoma and does not expressin tissues beside tumor. Positive rate in breast infiltrative ductal carcinoma is 54.17%. The differences between two groups are statistically significant (χ2=20.22, P=0.00).Positive rates with patients under(≤)50and over(>)50are69.35%and37.93%respectively. The differences between two groups arestatistically significant (χ2=11.92,P<0.05). VEGF-C positive rates of groupswithout lymph node metastasis and groups with lymph node metastasis are38.18%and67.69%respectively. The differences between two groups arestatistically significant(χ2=10.45,P<0.05).Positive rates of VEGF-C in breastcancer histological grades I、Ⅱ、III are42.86%、44.74%、75.68%respectively,the differences of which are statistically significant (P<0.05) and positivelycorrelated to histological grades of tumors (r=0.27, P<0.05). Expression ofVEGF-C has not been found to be related to tumor size.3. Correlation of expressions of Ki-67and VEGF-C in breast infiltrativeductal carcinoma of120cases with breast infiltrative ductal carcinoma,40casesare positive in both indexes,29cases are negative in both indexes,25cases arepositive in VEGF-C and negative in Ki-67,26cases are negative in VEGF-C andpositive in Ki-67. Correlation in expressions of VEGF-C and Ki-67has not beenfound(χ2=2.45, P>0.05).Conclusion1. A sharp rise of Ki-67protein expression in breast infiltrative ductalcarcinoma tissue was found and positively related to histological grade oftumors, indicating that the stronger proliferation activity is, the higher grademalignancy is.2. A sharp rise of VEGF-C protein expression in breast infiltrative ductalcarcinoma tissue was found and closely related to histological grades, lymphnode metastasis and patients’ age, indicating that ability to promote lymphatichyperplasia more strong, the higher grade malignancy is and metastasize moreeasily.3. No clear correlation between expressions of Ki-67and VEGF-C in breastinfiltrative ductal carcinoma has been found, indicating that enhancedproliferation activity and to promote the formation of lymphatic vessels are through different mechanisms of action.
