Objective:To study the expression of Interleukin-27(IL-27)、IL-35and their common subunit Epstein-Barr virus-induced gene3(EBI3) in patients of psoriasis vulgaris (PV), vitiligo and healthy control.Methods:Select peripheral blood from62cases of PV patients,with22cases in progressive,20cases in static, and20cases in paracmasis,120cases of vitiligo, with55patients in progressive and65patients in static, together with30healthy controls. Competitive inhibition enzyme linked immunosorbent assay was used to detect the levels of serum IL-27and IL-35, and SYBR Green I real-time RT-PCR was performed to measure the mRNA expression of EBI3in peripheral blood mononuclear cell (PBMC).Results:1. The expression of IL-27, IL-35and EBI3mRNA in peripheral blood in PV.1.1The levels of serum IL-27were significant higher in progressive(226.72±71.32pg/ml), static(160.80±20.02pg/ml) and paracmasis(181.93±27.36pg/ml) of PV than in healthy controls(138.66±33.93pg/ml)(P<0.01,<0.05,<0.01, respectively). There were increased significantly in progressive and paracmasis compered to static of PV (P<0.01,<0.05, respectively). The expression of IL-27in progressive were higher than in progressive, but there was no significants (P>0.05).1.2The expression of serum IL-35were significant higher in paracmasis (355.78±49.82pg/ml) of PV than in progressive(309.36±36.10pg/ml), static(299.14±31.98pg/ml) and healthy control (292.43±36.26pg/ml), all P<0.01. there were no significants between the group in progressive, static of PV and healthy control(p>0.05).1.3The PBMCs expression of EBI3mRNA in progressive, static and paracmasis of PV were20.15,16.17and19.74times higher than in healthy controls.2. The expression of IL-27, IL-35and EBI3mRNA in peripheral blood in vitiligo.2.1The levels of serum IL-27were significant higher in progressive (184.11± 50.70pg/ml) and in static (167.74±39.84pg/ml) of vitiligo than in healthy controls(138.66±33.93pg/ml)(all P<0.01). There were significant increase in progressive compered to static of vitiligo (P<0.05).2.2The expression of serum IL-35in progressive(348.69±36.26pg/ml) and static(16.01±53.68pg/ml) were significant higher than in healthy control(292.44±36.26pg/ml)(all P<0.01), There were significant increase in progressive compered to static of vitiligo (P<0.05).2.3The expression of serum IL-27in vitiligo focal, vulgaris, acrofacial and universal were174.57±43.15pg/ml,174.95±53.73pg/ml,166.30±27.40pg/ml,194.38±59.73pg/ml respectively. They were significant higher than in healthy control, all P<0.01, however, there were no significant different between themselves, all P>0.05.2.4The expression of serum IL-35in vitiligo focal, vulgaris, acrofacial and universal were322.72±65.18pg/ml,337.41±50.82pg/ml,328.04±33.42pg/ml,345.57±79.05pg/ml respectively. They were significant higher than in healthy control, all P<0.01, however, there were no significant different between themselves, all P>0.05.2.5The PBMCs expression of EBI3mRNA in progressive and static of vitiligo were11.69and7.48times higher than in healthy controls.Conclusion:1:IL-27, IL-35and EBI3mRNA may be involved in the pathogenesis of psoriasis vulgaris. IL-27and EBI3were likely play a pro-inflammatory and anti-inflammatory role in the course of the disease. IL-35had much higher levels in paracmasis, suggesting that it may promote psoriasis subside.2:IL-27, IL-35and EBI3mRNA in vitiligo early expression was significantly increased, suggesting that it may be involved in the pathogenesis of vitiligo disease, and the parting was not significantly related.
