Objective:This study used acute lung injury(ALI) model mice induced by lipopolysa-ccharide as the research object,intervened with the Qingfei jiedu decoction,Throughdetermining the levels of the mice serum inflammatory factor IL-1β and CRP,andobserving the morphological changes of lungs in mice,evaluating the anti-inflammato-ry effect of the Qingfei jiedu decoction on acute lung injury model in mice.Methods:Sixty healthy BALB/c mice were randomly divided into5groups: normalgroup, model group, the Qingfei jiedu decoction low-dose group,the Qingfei jiedudecoction high-dose group,dexamethasone group,each group of12.Above groups,inaddition to the normal group,were modeled with LPS by intranasal administration.Aft-er2hours modeling the mice of normal group and model group were given normalsaline l0ml/kg by gavage once per day;Qingfeijiedu decoction32.5g/kg,19.5g/kg wereadministered respectively to high-dose group and low-dose group by gavage once aday;the dexamethasone group were given dexamethasone1mg/kg by gavage once aday.The five groups were all administered for3days.Before modeling and duringtreatment, measure the ear temperature in mice at9am.After the last drug to kill themice and get the blood. Observe the appearance of their lungs and Weigh the bodyweight and lung weight,calculate the lung index.After HE staining,we observed themorphological changes of lungs with light microscopy,and to score.To determine thelevels of the serum IL-1β and CRP by ELlSA.Results:(1)Observation of the general condition:there were2deaths during theexperiment; the mice of normal control group were lively and active,their furs werebright,and they have eating normally;the model group appears reduced activity,Curlyfurs and no gloss,poor mental,lack of appetite;the Qingfei jiedu decoction high-dosegroup and low-dose group is better than the model group at activity,the glossiness offurs,mental state and diet.The general condition of dexamethasone group were similarto the Qingfei jiedu decoction groups.(2)Comparison of lung index:the lung index of the model group has a very significant difference with normal group(P<0.01); the lungindex of the Qingfei jiedu decoction low-dose group,the Qingfei jiedu decoctionhigh-dose group and dexamethasone group has difference with the model group(P<0.05).However,there was no significant difference between the Qingfei jiedudecoction low-dose group, the Qingfei jiedu decoction high-dose group and dexamet-hasone group.The difference between the Qingfei jiedu decoction low-dose group andhigh-dose group had no statistics significance(P>0.05).(3)Comparison of plasmaIL-1β and CRP:the concentration of plasma IL-1β in model group was higher than thatin normal group(P<0.01);compared with model group,the concentration of plasmaIL-1β in the Qingfei jiedu decoction low-dose group, the Qingfei jiedu decoctionhigh-dose group and dexamethasone group was lower(P<0.05).however,there was nosignificant difference between the Qingfei jiedu decoction low-dose group andhigh-dose group; the concentration of plasma CRP in model group was higher thanthat in normal group(P<0.01).compared with model group,the concentration of plasmaCRP in the Qingfei jiedu decoction low-dose grou(the Qingfei jiedu decoctionhigh-dose group and dexamethasone group was lower(P<0.05).compared withdexamethasone group,the concentration of plasma CRP in the Qingfei jiedu decoctionlow-dose group and high-dose group was higher,but had no statistics significance(P>0.05).There was no significant difference between the Qingfei jiedu decoctionlow-dose group and high-dose group (P>0.05).(4)Results of pulmonary tissuepathology and pathological integral: the normal group mice’lung tissue structure wereclear, alveolar were intact, alveolar interval were uniform,and have no inflammatorycells.There is visible diffuse thickening of alveolar interval and a large number ofinflammatory cells(Necrotic neutrophil and mononuclear lymphocyte cells)in themodel group;there is part of the alveolar interval thickening and a small amount ofinflammatory cells(mononuclear lymphocyte cells) in the Qingfei jiedu decoctionlow-dose group,the Qingfei jiedu decoction high-dose group and dexamethasonegroup.The pathological integral was higher in model group than that in normal group(P<0.01);compared with model group,the pathological integral in the Qingfei jiedudecoction low-dose group,the Qingfei jiedu decoction high-dose group and dexameth-asone group was lower(P<0.01),however,there was no significant difference betweenthe Qingfei jiedu decoction low-dose group and high-dose group(P>0.01),There wasno significant difference between the Qingfei jiedu decoction low-dose group andhigh-dose group(P>0.05). Conclusions:(1)The Qingfei jiedu decoction can reduce the concentration of plasmaIL-1β and CRP,and reduce the lung index.There was no difference in the efficacybetween high and low dose;(2)The Qingfei jiedu decoction can reverse the change ofpulmonary tissue pathology and reduce the lung semi-quantitative score.Either high orlow dose had no difference;(3)There was no difference in the curative effect betweenThe Qingfei jiedu decoction and dexamethasone in reducing the concentration ofplasma IL-1β and CRP, reducing the lung index,and reversing the change ofpulmonary tissue pathology.
