| Objective:This study basis of the early research of our team, included innovative processingmethod, stomach intestine fluid metabolism, metabolic product, metabolomics, theantioxidant activity of compound, the activity of blacking the hair and the dissolutionrate of compound formula, to continue in-depth study of processing theory, clearingthe difference as well as the similarities pharmacological effects after processing,providing evidence of clinical application.Methods:1, Using RP-HPLC method to determinate the content of freeze-drying and sevenkinds of traditional processing method of Polygonum Multiflorum Radix.2, Using RP-HPLC method to determinate the content of the reaction products by theartificial stomach intestinal juice of Polygonum Multiflorum Radix.3, After proper treatment to Polygonum Multiflorum Radix, injecting to LC-MSsystem, and fully tested its chemical composition.4, lavaging SD rats and then collecting its metabolites, using LC-MS to detect thechange rule of elements in the body metabolism.5, Putting Polygonum Multiflorum Radix and boiled Polygonum Multiflorum Radixrespectively into Jingwucapsule, using DPPH, FRAP method, hydroxyl free radicalclearance method to determinate its antioxidant activity, Lavaging male healthC57BL6/N mice, measuring the length of hair in mice and melanin content,detecting the index of mice skin of FGF-7, MC1R, VEGF, MSH, HGF, IGF-1, SHH, Using RP-HPLC method to determinate the content of the compound formula.Results:1, By contrast,had found TSG content in the lyophilized product was the highestwhile lowest in the steamed products; Emodin content was highest in black beanproducts while lowest in the lyophilized product. Physion content was highest in thecrude drug while lowest in the lyophilized product2, The raw Polygonum Multiflorum Radix after making compound formula threecomponent content all had varying degrees of decline, the decline range was lesser,styrene with two was7.6%;Free emodin by64%;Free emodin methyl ether by129%.Polygonum Multiflorum Radix after steamed,the content of TSG in slightlyincrease (7.7%), other two ingredients showed a trend of decline (free emodin by51.7%; free emodin methyl ether by152%3, From the heat maps of LC-MS, a total of1105compounds for clustering analysis,PCA method of raw and steamed products classification was good, compared withraw Polygonum Multiflorum Radix, processing characteristic peak changedsignificantly after a total of344,97of the characteristic peak levels rise,247characteristic peaks were decreased.PLS-DA VIP chart shows that the VIP valuegreater than1characteristic peak, a total of344created after97characteristic peaklevels rise, accounts for about28%, to247characteristic peak content was lower,about72%.4,In the artificial gastric juice: compared with raw Polygonum Multiflorum Radixgroup, the steamed one content of TSG,free emodin, free emodin methyl etherdissolution rate was higher, the dissolution rate difference of TSG was small, freeemodin, free emodin methyl ether dissolution rate of differences.In the artificialintestinal juice: the raw one,s dissolution rate of the three components were all higherthan the steamed one.5, The raw one antioxidant capacity was highter than processed, and from theperspective of the numerical value of TSG, TSG showed strong antioxidant capacity.The raw made into compound formula,its anti-oxidation ability was strongerthan the compound formula made by steamed one.6,The raw capsule and steamed capsule both has the role of promoting hair growth,and has been a trend of time, with the passage of time, the hair longer and longer, andvolatility was becoming more and more small, but the steamed capsule group workingboth speed and length of hair hair growth effect were better than raw capsule group.Analysis found that raw capsule group work more quickly than steamed capsule group,but persistent was short; by contrasted, the steamed capsule group work slower but itcan continue a long time, and showed higher with the extension of growing hair index.It could be seen from the hair melanin measurements, the steamed capsule couldsignificantly improve the melanin content in the hair.7, Using the liquid mass spectrometry analysis, found three composition changes aftermetabolites, TSG and emodin methyl ether and its related metabolic components werenot detected in the urine and feces. Emodin prototype components were detected inthe urine but not detected in the feces.Conclusion:1, Freeze drying method could better keep the TSG of fresh Polygonum MultiflorumRadix.2, TSG of Polygonum Multiflorum Radix approved by processing, the compatibilityof stomach and intestinal juice, the content of basic remain stable, but the content ofemodin and emodin methyl ether is vary considerably.3, TSG of Polygonum Multiflorum Radix antioxidant activity and had a greater influ-ence on the activity in hair grow and been black.4, When develop and utilize Polygonum Multiflorum Radix as antioxidant and on theactivity in hair grow and been black should avoid processing and influence of artifice-al gastric juice or artificial intestinal juice by using damage to TSG. |
