Objective:By establishing the LEASO model in rabbits,to observing the preventive effectof GSPE on LEASO, and exploring its mechanism of action. Detective the indicatorsof ox-LDL, MDA, SOD, NO.Explore the prevention mechanism of GSPE on LEASO.Methods:30male New Zealand rabbits were randomly divided into three groups (control,LEASO and GSPE group), with10rabbits in each group.the control group fed with o-ridinary food,the model and GSPE group fed with high fat diet for a week. Then,wehave an operation on LEASO and GSPE group(cut the right femoral artery and use th-e2F Fogarty balloon catheter to femoral artery intima). Blood samples were drawn fr-om the ear vein of rabbits at different time points(4,8,12weekend of the experiment-t).The blood samples were analyzed for ox-LDL, MDA, SOD and NO.At12weeken-d,to observe the right femoral artery blood flow by color doppler ultrasound and digi-tal subtraction angiography (DSA).Aorta and the right arteria femoralis were excisedand morphol-ogically observed through HE staining.Result:1.Weight:During the12th week, the weight increaseed more significantly in theLEASO and GSPE group than the control group.(P<0.01).2.Total cholesterol(TC) test results: At the12th weekend, the serum TCincreaseed more significantly in the LEASO and GSPE group than the control group.(P<0.01);LEASO group compared with GSPE group,the serum TC had no significantdifference(P>0.05).3.Oxidized low-density lipoprotein(ox-LDL) and malonald-ehyde(MDA) testresults: At the12th weekend, the serum ox-LDL increased more signifycantly in theLEOAD and GSPE group than the control group(P﹤0.01). In the LEASO group,ox-LDL was higher than GSPE group, respectively(P﹤0.01). 4.Malonald-ehyde(MDA),superoxide dismutase(SOD) and nitric oxide(NO)test results: At the12th weekend, the serum MDA increased more signifycantly in theLEOAD and GSPE group than the control group (P﹤0.01). In the LEASO group,MDA were higher than GSPE group, respectively(P﹤0.01).The serum SOD and NOdecreased more significantly in the LEASO and GSPE group than the controlgroup,all P﹤0.05. In the GSPE group,SOD and NO were lesser than LEASO group,respectively(P﹤0.01).5.General condition of right lower extremity: During the process of experiment,thelower extremities of rabbits in CON and GSPE group are normal;At5thweeked,threeof the rabbits in LEASO group appeared claudication in right lower extremity.And at8thweekend,one of three rabbits appeared gangrene in right lower extremity.6.Detection of color doppler ultrasound:The right femoral artery blood flow ofLEASO group is worse than GSPE group.7.Detection of digital subtraction angiography (DSA):in LEASO group, theterminal of right femoral artery development wasn’t satisfied.While the GSPE wasmuch better.8.Morphologically, the progress of atherosclerosis was obviously alleviated byGSPE while the GSPE group was better than the LEASO group.Conclusion:1.GSPE can significantly alleviate the development of atherosclerosis. It can alsopromote the formation of collateral circulate-on and improve the blood supply to thedistal limb effectively2.GSPE has protective effect on LEASO,its mechanisms may be related to inhib-iting the formation of free radicals and the lipid peroxidation... |
