Objective:1. This study uses immunohistochemical methods to mark the lymphatic duct of tongue squamous carcinoma lesion and lymph node metastasis, and calculate the lymphatic vessel density to observe the expression of CEACAM1in intratumoral lymphatic vessel and peritumoral lymphatic vessel. Then analyze the function of CEACAM1and its acceptor in the developing and lymph node metastasis of tongue cancer.2. To establish an orthotropic model of human tongue squamous cell carcinoma in nude mouse, simulate human body environment, observe the tumors cell biological behavior and condition of lymph node metastasis in vivo. To mark the lymphatic duct of tongue squamous carcinoma lesion and lymph node metastasis by immunohistochemical methods, and calculate the lymphatic vessel density to observe the expression of CEACAM1in intratumoral lymphatic vessel and peritumoral lymphatic vessel. Then analyze the function of CEACAM1and its acceptor in the developing and lymph node metastasis of tongue cancer.Methods:1. Detection of72cases of tongue squamous cell carcinoma lesion by immuohistochemical method, with43metastasis positive patients(male21cases, female19cases) and29metastasis negative patients(male17cases, female12cases). Using rabbit anti-human CEACAM1marker to mark lymphatic,and count the lymphangiogenesis density of intratumoral and peritumoral. Data is analyzed with SPSS20.0.2. The Tca83cell are injected into the left margin of tongue in nude mouse. To Check and record the weight, the condition of tumor growth, the length and width of tumor every other day. Mouse were sacrified when they died or their weight were less than25%of initial data. Then dissect their tongue, lymph nodes. Using rabbit anti-human CEACAM1marker to Mark lymphatic, and count the lymphangiogenesis density of intratumoral and peritumoral. Data is analyzed with SPSS20.0. Results:1. The intratumoral exist isolated、neonatal、incomplete、tubulous structure, not with the surrounding host lymphatic endothelial cell gap is communicated, the tubulous structue can be seen nore, luminal narrow; the number of peritumoral lymphatic vessels,which are larger and dilated, is more. The peritumoral lymphatic vessels density with lymph node metastasis(16.748±3.070) was significantly higher than the intratumoral lymphatic vessels density(8.662±3.214, P<0.01).2. Tumor was formed in6th-12th day after injected tumor cell. Tumor formation rate was77.14%, the result of HE and Immunohisochemical showed cervical metastasis rate was34.29%. Both intratumoral and peritumoral have positive expression, showed lymphatic irregular shape, thin wall, can be in open state, cavity does not contain red blood cells, and blood vessels are obviously different, sometimes with jelly lymph; also can behave for lumen collapse, atresia were cord-like, even a single endothelial cells; sometimes visible lumen is not complete.he peritumoral lymphatic vessels density with lymph node metastasis(5.488±2.114) was significantly higher than the intratumoral lymphatic vessels density(3.026±1.470, P<0.01).Conclusions:1.(1) CEACAM1is specific and sensitive to endothelial cells of lymphatics, so it can be applied to the lymphatic research.(2) The lymphangion genesis, both in peritumoral and intratumoral tissues, increase the possibility of lymphatic metastasis, and play a great role in the growth and metastasis of tongue cancer.2. The establishment of a orthotropic mode human tongue squamous cell carcinoma in nude mouse was successful. It supplied an important experiment medium in studying tongue cancer development and metastasis. The study, which compare the peritumoral lymphatic vessel density and intratumoral lymphatic vessel density, suggests that lymphangion genesis may accelerat the process of lymphatic matastasis.
