| Objective:The Y chromosome could be continued in paternal heredity, changed steadly and gradually in paternal heredity, and establish parallel correspondence with the surname followed by the paternal heredity via molecular coalition analysis of the Y-SNPs and Y-STR. theoretically, each surname should have its own unique core haplogroup, The samples from the surname Duan were selected to study, including12Y-SNPs with high polymorphism in Eastern Asia and23Y-STRs from the PP-Y23kit. The difference of haplogroups frequency between the samples from surname Duan population and unrelated male population is compared. All this will be useful for exploring the correlation between surname and Y chromosome in forensic applications studies.Methods:102unrelated male samples who share the same surname Duan and330unrelated male samples and255unrelated male samples share the same surname Kong obtained randomly were collected. Chelex-100and the phenol-chloroform method were adopted to extract DNA. Two multiplex genotyping assays and SNaPshot assay were used to analysis all samples which allows to obtain the Y haplogroups.23Y-STRs were further typed for the surname Duan population samples. The software package Arlequin3.5.1.2and the program SPSS17.0were used for data statistical analysis. Results:13haplogroups were observed including C-M130, D-M174, K-R*-M9, O*-M175,O1a-M119, O2al-M95, O3*-M122, O3a*-M324, O3a2*-P201, O3a2b-M7, O3a2cl*-M134, O3a2cla-M117and Non-M9. The haplogroup frequenc y of the Duan populations is C-M130(0.0990)ã€D-M174(0.0693)ã€K-R*-M9(0.0495)ã€O*-M175(0.0792),O1a-M119(0.0594)ã€O2al-M95(0.1386)ã€O3*-M122(0.0396)ã€O3a*-M324(0.1881)ã€O3a2*-P201(0.0495)〠O3a2b-M7(0.0099)ã€O3a2cl*-M134(0.0792)ã€O3a2cla-M117(0.1089)ã€éžM9(0.0297).102haplotypes in Duan population defined by23Y-STRs1ocus were obtained and the haplotype diversity is1. Compared the Y-STR mut ation rates among all locus,. DYS19appears allele10(0.0099) and12(0.0297) which were not reported in East Asia; DYS389II had two deletions of23and33; DYS390had two deletions of21and28; The polymorphism of DYS391i s poor, a higher rate of different mutations at this locus in East Asian populati ons,etc. The typical haplotype is18-12-20-28-15-10-21-12-11-10-14-18-20-23-12-14-11-12-17-15-11Conclusions:The samples were restricted by the region the nationality and the surname which would be a model used as a Y chromosome and the surname of parallel transmission..It would explore the association between Y chromosom e and name. Combined with Y-SNP and Y-STR analysysing showed that the Duan population experienced gene drift which has its own origins and develop ment. |
