Background Acute kidney injury is a severe crisis in clinical works. A variety of reasons can cause acute kidney injury, such as severe infections and drugs, et al. Although with improvement of treatment technology, acute kidney injury mortality has declined, but the mortality of intensived care patients combined AKI still amounte to about65%. The treatment of AKI is mainly renal replacement therapy currently. The treatment of adipose-derived mesenchymal stem cells has become one of the hot researchs for Nephrology workers recently. Experimental studies have shown that ADMSCs has a role in the treatment of acute kidney injury, and it have many advantages, such as its rich sources, easily isolation and culture, low immunogenicity, et al. however,the mechanisms is still not precise.Objective1. To evaluate ADMSCs’ effort on the cure of acute kidney hurt by observing subjects with kidney hurt.2.To probe into the possible mechanism to cure AKI through ADMSCs by determining related factors, so as to provide experimental example of clinical application of ADMSCs to cure AKI.Methods1. Firstly group the30male KM rats into three ones on average, namely normal control group, model group and treatment group. Secondly set up the model of kidney hurt by injecting cis-platinum under enterocoelia. 2. then injecting suspension of ADMSCs into caudal vein of those of treatment group in12hours, the same equivalent normal saline into caudal vein of those of treatment group in12hours.3. After seven days put the rats to death by injecting cis-platinum. The condition of kidney hurt can be judged by observing the morphological changes of their nephridial tissue and measuring the numbers of BUN and Scr. The conveying of NGAL, c-KIT, SCF can be measured by the method of IHC.Results1. Compared with the model group, the number of BUN、Scr and NGAL of kidney hurt of are greatly decreased and so it is with the possibility of ATN(P<0.05).2. Compared with the model group, c-KIT、SCF of the treatment group are increased greatly, the same result can be attained when compared with the normal control group(P<0.05).Conclusions1. Exogenous ADMSCs can enhance the function of repair of the injured renal tubular epithelial cell and also can decrease the number of markers of kidney injury.2. In the model of AKI, exogenous ADMSCs can enhance the function of repairing of the injured renal tubular epithelial cell and Promoting the Proliferation,differentiation of the tubular epithelial cell to protect the injured kidney by the paracrine or its secreting of SCF、c-KIT and its activating of SCF/c-KIT.
