BackgroundParkinson’s disease(PD) is common in the elderly with chronic progressive neurodegenerative disease.Currently the majority treatment of Parkinson’s relieve symptoms and improve the quality of life of patients based. Currently, there is no definitive diagnosis of early objective indicators, and there is no effective way to prevent the occurrence and progress of PD.Many studies indicate that glial cell line-derived neurotrophic factor on neurons in the brain has a protective effect, its nutritional effect on neurons must pass transmembrane information transfer effect, namely to achieve receptor-mediated and receptor-mediated GFRas(GFRal,GFRa2,GFRa4) in guide the process of the biological effects of GDNF plays a key role, accordingly minocycline of protective effect on PD also observed.Objective(1)To understand the distribution and variation of GFRas in PD rat model’s substantia nigra and the striatum on different time points to further suggest the relevance of the GFRas and the PD.(2)Application of minocycline role in the PD rat model, to further understand the change of GFRas on different time points, in order to explore the protective effects of minocycline on PD.Method(1) The rats were randomly divided into four groups:normal group, sham-operation group, the PD model group and minocycline group.(2)Using the subcutaneous injection of Apo morphine induced the rotation behavior to evaluate if the PD model successful.Application of mNSS scoring scale to comprehensively evaluate the changes of rat movement, sensation, reflection and balance function.(3)Using the SP immumohistchemistry to detect the expression of GFRas in substantia nigra and striatum respectively of all model rats. To discuss the distribution and variation of GFRas in substantia nigra and striatum of rats model on different time points and the protective effects of minocycline on PD.Result1The results of behavior detectionThere is no obviously abnormal behavior change on the normal group and sham-operated rats,the mNSS ratings are0point. The postoperative PD model group and minocycline group rats had tremors, bradykinesia and other typical clinical manifestations of PD,and and acts of resisting arrest appears weakened, arched and other symptoms. Thirty-three PD model rats mNSS score between13to18points, it belongs to the severe damage;thirty-one minocycline group rats mNSS score between7to12, it belongs to the moderate damage.2The results of the SP immumohistchemistry analysis2.1The result of GFRα1in the substantia nigra and striatum(1)From the same time point:the level of GFRal in the PD rat group substantia nigra and striatum was lower than the blank group and the sham operation group. The difference was statistically significant (P<0.05). Compared with the PD group, the level of GFRal in the minocycline group increased, the difference was statistically significant (P<0.05).(2)From the different time point:the GFRal expression of the PD group showed a declining trend and GFRal expression of minocycline group showed a increasing trend, the difference of two groups at each time point were no statistically significant(P>0.05).2.2The result of GFRa2in the substantia nigra and striatum(1)From the same time point:the levels of GFRa2between normal group and sham-operation group were no significant difference (P>0.05). The level of GFRa2in the PD rat group substantia nigra and striatum was lower than thenormal group and the sham-operation group (P<0.05), which was statistically significant.Compared with the PD group, the level of GFRa2of minocycline group was higher (P<0.05), which was statistically significant.(2)From the different time point:the levels of GFRa2reached to the lowest peak at the one day point;the GFRa2relative expression of the PD group showed a declining trend and GFRa2relative expression of minocycline group showed a increasing trend, the difference of two groups at each time point were no statistically significant(P<0.05).2.3The result of GFRa4in the substantia nigra and striatum(1)From the same time point:the levels of GFRa4between normal group and sham-operation group were no significant difference (P>0.05). The level of GFRa4in the PD rat group substantia nigra and striatum was lower than the normal group and the sham-operation group (P<0.05), which was statistically significant.Compared with the PD group, the level of GFRa4of minocycline group was higher (P<0.05), which was statistically significant.(2)From the different time point:the GFRa4relative expression of the PD group showed a declining trend;GFRa4relative expression of minocycline group showed a increasing trend and reached to the highest peak at the fourteen day point, the difference of two groups at each time point were no statistically significant(P>0.05).ConclusionWith time, the expression level of PD model group GFRα1,GFRa2,GFRa4decreased; minocycline upregulate the expression level of GFRal, GFRa2,GFRa4and thus play the nutritional support, protection,injury repair and biological effects on PD dopaminergic neurons.
