Objective:After intranasal administration of recombinant human erythropoietin (rhEPO) in acute cerebral ischemia in rats,to investigate the neuroprotective effect of it by the expression of neuroglobin (Ngb) and tumor necrosis factor a (TNF-a) in the rat brain. Provide experimental evidence for intranasal administration of rhEPO in clinical.Methods:Seventy-two healthy male SD rats were randomly divided into three groups:control group (n=24), model group(n=24) and rhEPO treatment group(n=24). We adopted the suture method to make permanent middle cerebral artery occlusion model. rhEPO treatment group was given48u rhEPO via intranasal administration after ischemia for1hours, control group and model group were given the same volume saline at the time.All rats were decapitated after ischemia for24hours, the water contents in the brain tissues were calculated by dry-wet weight method, the infarction volume was measured by TTC staining (2,3,5-triphenyl tetrazolium chloride), the brain tissue form was observed by HE staining, and the expression of neuroglobin (Ngb) and tumor necrosis factor a (TNF-a)were detected by immunohistochemistry.Results:(1) The water contents in the brain tissuesCompared with the control group, the brain tissue water content of model group increased.Compared with the model group, the brain tissue water content of rhEPO treatment group diminished. The difference are Statistically significant.(2) The infarction volume in the brain tissuesThe control group had no volume of infarction in groups. Compared with the model group, the volume of infarction of rhEPO group reduced, The difference are Statistically significant.(3) immunohistochemistryThe expression of Ngb and TNF-a can be seen in every groups. Compared with the control group, the expression of Ngb and TNF-a of model group showed increasing,.Compared with the model group,the expression of TNF-a in rhEPO group decreased, the expression of Ngb in rhEPO group increased, The difference are Statistically significant. Conclusion:(1) Intranasal administration of rhEPO is an effective drug application method, The intranasal administration of rhEPO after1h of ischemia in rats decreased the volume of after24h infarction and reduced the brain tissue water content.(2)The intranasal administration of rhEPO after1h of ischemia in rats was of certain neuroprotective effect, its mechanism might be related to the reduction of expression of TNF-a as well as the elevation of expression of Ngb.
