Micromorphology Of Endoplasmic Reticulum And Expression Of GRP78in The Bladder Detrusor Recontruction Of Male Rat Model Of Partial Bladder Outlet Obstruction

ObjectiveTo observe the micromorphology of bladder detrusor and the expression of biomarker of endoplasmic reticulum stress(glucose-regulated protein78,GRP78),in the bladder recontruction of male rat model of partial bladder outlet obstruction.our aim is to explore endoplasmic reticulum stress playing a role in the bladde reconstruction.MethodsFourty male Wistar rats were divided into control group(sham group,10),2-weeks of obstruction(15) and4-weeks of obstruction(15).we observed the micro structrue of bladder detrusor smooth cells using transmission electron microscopy.According to immunohistochemical and real-time PCR methods,we respectively detected expression of proteins and mRNA of GRP78.ResultsThe expression of GRP78protein was (3.37±0.38)%,(51.22±0.27)%,(27.02±1.85)%in sham-operated group,2weeks of obstruction and4weeks of obstruction, respectively. There was significant difference between2weeks of obstruction and4weeks of obstruction group (P<0.01). The difference between sham-operated group and obstruction group was significant (P<0.01). The expression of GRP78mRNA was (22.21±1.58),(30.62±2.49),(24.52±2.24),There was significant difference between sham-operated group and2weeks of obstruction group(P<0.01).The difference between2weeks of obstruction group and4weeks of obstruction group was significant (P<0.01).ConclusionsIt is feasible method that perineal approahc.In the process of bladder reconstruction after partial bladder outlet obstruction,there is a evidently changes in the morpholopy of bladder detrusor cells endoplascim reticulum, with obviously expression of protein and mRNA of GRP78.This suggested that endoplascim reticulum stress play a role in the process of bladder reconstruction.
