| Objective:To study the inhibition, cytotoxicity and cell morphology and the proportion of cancer stem cells of salinomycin effected on breast cancer MCF-7cells in vitro,Discuss the possible mechanism of these effects.Methods:1,Used8different concentrations of Salinomycin(100ã€50ug/ml etc) effect on breast cancer MCF-7cells, Docetaxel for the control, detected the half-effect concentration(IC50ug/ml) by Cell Counting Kit8.2,The human breast cancer MCF-7cells were cultured in adherent and serum-free suspension culture method, The expression of CD44+CD24-/low was detected by Flow Cytometric analysis. We observed their ability to formation of mammospheres by serum-free suspension culture method, the effect of salinomycin, docetaxel on these mammospheres.3,Proportion of CD44+CD24-/low cells in breast cancer MCF-7cells that treated with salinomycin, docetaxel were assessed by Flow Cytometry.4,The protein levels of P-glycoprotein were detected by Western blot.Results:We cultured mammospheres in which expression of CD44antigen was significantly higher,CD24antigen was lower or negative. The inhibition to proliferation of breast cancer MCF-7cells and cancer stem cells by salinomycin was significantly stronger than docetaxel or Salinomycin restrained the ability of mammospheres formation and downregulated the level of P-glycoprotein.Conclusions:The breast cancer MCF-7cells and cancer stem cells were more sensitive to salinomycin than docetaxel. |
