| Objective:To investigate the relationship between the polymorphisms of CXCL10-201A/G with the susceptibility of enterovirus71encephalitis, and explore the risk factors of EV71encephalitis.Method:Blood samples were collected from174children with EV71infection confirmed and227normal control cases without EV71infection. Genomic DNA of white blood cell were extracted, then polymerase chain reaction with sequence-specific primers (PCR-SSP) system were used to determine genotype for the A/G polymorphism at-201position of CXCL10gene in encephalitis group and in control group.Results:1. The frequencies of genotype GGã€GA and AA in the position of CXCL10-201A/G were56.9%,34.48and8.62%in174case samples, while55.07%,39.65and5.28%in227control samples. No obvious significances between EV71infected group and control group were found (P>0.05).In case samples, the frequencies of genotype GG〠GA and AA in103EV71mild cases of were62.14%,33.98%and3.88%, while in71severe cases were53.52%,30.99%and15.49%. Children who carry AA have a higher risk to develop into severe encephalitis (X2=6.891,P=0.032)2. The frequencies of allele G and A in the position of CXCL10-201A/G were74.14%and25.86%in174case samples, while74.89%and25.11%in227control samples. No obvious significances between EV71infected group and control group were found (P>0.05). In case samples, the frequencies of allele G and A in103EV71mild cases were79.13%and20.87, while in71severe EV71cases were69.10%and30.99%. Children who carry allele A have a1.702times higher risk to develop into severe encephalitis (OR=1.702,95%CI=1.043-2.776)3. The information of71cases samples, including gender, age, temperature, convulsion, consciousness disorders, pathological signs, WBC, CRP, ALT, AST, CKMB and cranium MR, were recorded. The frequencies of genotype GGã€GA and AA showed significant differences in age, consciousness disorders and pathological signs, in other items we found no obvious differences. In laboratory examinations, for ALT, AST and children who carried AA genotype had a much higher level than other types, not in the rest items (P>0.05)4. In CSF test of71cases samples of encephalitis, children who carried A allele had a higher level of chlorides and cell counts than children carrying non A with no statistical significance (P=0.086åŠP=0.06), not in Glucose, Protein, Lymphocyte and Monocyte (P>0.05)Conclusion:The polymorphism of CXCL10-201A/G single nucleotide may correlate with EV71encephalitis; maybe CXCL10-201A/G GG is the protective genotype which protects children to keep away from EV71serious encephalitis. |
