| Objective: To explore the effect and Mechanism of autophagy specific gene beclin-1on gastric cancer cell SGC7901in Vasculogenic mimicry forming abilityMethods: Based on the beclin-1, an autophagy specific genes, we establishautophagy inhibited group, negative control group and blank group.Then we investigatedthe different VM forming ability of that three groups, and detected the gene expressing(c-myc,oct-3/4,sox-2,notch-1,MMP-2,9,E-cad), survival and invasive abilitychange correspondingly.Results: After3D culture in matrigel, gastric cancer cell SGC7901can form VM andthe expression of autophagy specific gene beclin-1was increased (P<0.05).Howeverwhen we knockdown the expression of beclin-1by RNA interfered, VM can’t form: VMnumblers of blank group, negative control group and beclin-1inhibited group were(37.6±4.5,37.8±1.9,15.4±1.1, P<0.05). By the more, the expression ofPluripotencygene (c-myc) decreased in beclin-1inhibited group than blank group and negativecontrol group,(1,3366.3±109.2,7475±174.1,7433.3±150,P<0.05). transwell showed thatthe invasiveness ability and survival ability in starvation environment of beclin-1inhibitedgroup was reduced than other two group.Conclusion: In the forming of VM, the stable expression of genes was indispensable.under the adverse environment, autophagy can be activated to maintain the stability ofgenes expression, Finally promote consistion VM,which can convey oxygen and nutrientsinto the central of tumor tissue, at the same time, provide transfer channels to facilitatecancer cell to escape from the stress environment. |
