ã€Objective】The endothelial cells of hemangioma were cultured by explant combined withtrypsin-digestion technique. The cells were injected subcutaneously into nude mice toestablish a mouse model of hemangioma, given propranolol gel coated intervention,To investigate the effectiveness of propranolol gel inhibited the growth ofhemangiomas,to provide a theoretical basis for the treatment of hemangiomaspropranolol gel.ã€Methods】Fresh operative specimens were obtained from infantile hemangiomas.Theendothelia cells of IH were cultured by explant combined with trypsin-digestiontechnique.The purity of endothelial cells were examined by flow cytometry analysis.Immunohistochemical staining method was choosing to identify the cultured cells bydetected the expression of CD31antigen,CD31antigen and Factor VIII related antigen;The cultured cells was made into cell suspension as a density1×107cells/300μl, mixedwith200μl Matrigel was injected into the flank of the Balb c/nu nu nude mice,three and five days after injection, an additional1×107cells/200μl was injected again.Mice were divied into control and experimental groups. The other10normal mice,fifteen days after injection, experimental group were giving3%propranolol gel coatedon cell injection site,3times/d, the normal group were also giving3%propranolol gelat equivalent time,the control group were giving without any treatmenting. Changes ofthe tumor were observed for different periods. Compare the difference between the twogroups of tumor growth. ulceration, bleeding, damaging were also observed on the shinwhere were giving3%propranolol gel coated; At45days, the mice were killed andsamples were taken from the surviving tumors, blood plasma concentration detection,tumor tissue excised HE staining and immunohistochemistry, and examined by HE staning and Immunohistochemical staining for pathology.ã€Results】1.Endothelial cells can be seen after tissue culture3d, and cells covered bottom at16d;2.The expression of factor â…§ associated antigen, CD31, CD34was positive in theendothelial cells in vitro;3.Flow cytometer indicate the purity of hemangiomas cell was99.47%;4.In experimental group, tumor disappeared after cells were injected for30day, and theskin without bleeding, ulceration, damaging appear;5.In the control group, tumor slowly increasing after cells were injected for15day, andlargest at day45;6.HE staining showed a lot of angiogenesis, immunohistochemistry confirmed as thesource of human hemangioma.ã€Conclusions】1. Endothelial cells of hemangioma can be cultured by explant combined withTrypsin-digestion technique;2. Hemangioma model can be successfully established in nude mice;3. Propranolol inhibit the growth of hemangioma;4. Propranolol gel was effective, no significant adverse reactions in treatmenthemangioma. |
