| Objective:1. Whether magnesium supplement can improve the motor symptoms of6-OHDA inducedPD rats;2. Whether magnesium supplement can antagonist6-OHDA lesioning in striatum dopaminesystem;3. Gene expression of magnesium transporters SLC41A1, MagT1and CNNM2in6-OHDAinduced rat model of PD.Methods:1.8g of6-OHDA was injected into the right MFB by stereotaxic apparatus to get PD rats.2. The therapitic experiment beginning at2weeks post injection of6-OHDA. For treatmentgroups, we use3.6g/L MgSO47H2O to replace daily drinking water, and raise2weeksor4weeks, respectively. Two or4weeks later, evaluate the therapitic effect by APOinduced rotational test.3. Preventive experiments beginning at half an hour post6-OHDA injection, a dailyintraperitoneal injection of90mg/kg MgSO47H2O was used for treatment and last for1or2weeks respectively, then launch APO induced rotational test and curling test toevaluate the damage degree of the nigra and striatum dopamine system.4. Detecting magnesium transporters SLC41A1, MagT1and CNNM2mRNA expression instriatum of rats in all of the experimental groups by q-PCR.5. Detect magnesium transporters SLC41A1, MagT1and CNNM2protien expression instriatum.Results: 1. The theraputic experiment results showed that the APO induced total turns in30mindecreased significantly after magnesium treatment for2weeks and4weeks, and for2weeks, there are significant difference between PD/Mg group with a speed of312±47r/30min and PD group with a speed of447±53r/30min (p <0.05).2. The preventive experiment results show that the APO induced total turns in30mindecreased significantly after magnesium treatment for1week and2weeks. For1weeks ofmagnesium treatment, PD/Mg group with a speed of199±64r/30min vs PD group with280±62r/30min (p<0.05). For2weeks, PD/Mg group with a speed of238±44r/30min vsPD group with384±25r/30min (p <0.05). The curling test results show that the PD/Mggroup with a score12.2±2.6was lower than PD group with a score17.1±3.1(p<0.05).3. The results of Q-PCR and Western blot showed that SLC41A1, MagT1and CNNM2geneexpression in lesioned striatum of PD/1week groups and PD/2weeks was lower than that incontrol groups(p<0.05), but there were no sinificant difference in PD/4weeks vs controlgroups, PD/6weeks vs control, the expression of SLC41A1, MagT1, CNNM2mRNAwas significantly higher than that of control group (p <0.05). However, only the level ofSLC41A1protein expression was obviously higher than that of control group in the samecondition (p <0.05).Conclusions:1. Magnesium supplement can improve the motor symptoms in6-OHDA induced PD rats2. SLC41A1, MagT1, CNNM2may invovled in the6-OHDA induced cell damage progressby its suppression of gene expression. And its high expression in striatum of PD rats maybe induced by the candition that magnesium diffeciency in the PD rat brain.. |
