| ObjectiveTo detect the expression of ALDH3A1gene in different time of HCC (hepatocellularcarcinoma) recurrence/metastasis after radical resection (R/M≤6Monthsgroup, R/M6-12monthsgroup and NR/M group within two years). And explore the relationship betweenALDH3A1genes and the early recurrence/metastasis of large HCC recurrence andmetastasis combined with the corresponding eighteen clinicopathological indices.MethodsCancerous and noncancerous tissues form56large HCC pateints who had undergone surgeryat the liver center of the First Affiliated Hospital of Fujian Medical University of China fromJanuary2006to January2012were collected, including23cases in R/M≤6Monthsgroup,20cases in R/M6-12monthsgroup,and13cases in NR/M group within two years. Firstly, eighteenclinicopathological indice related to the early recurrence/metastasis of large HCC recurrenceand metastasis were analysed. And then using Real-time Quantitative PCR to detect themRNA expression of ALDH3A1in liver cancer tissues and adjacent tissues. Meanwhilecombined with clinicopathological factors to multivariate analysis the relationship betweenALDH3A1and the early recurrence/metastasis of large HCC.Results1. Univariate analysis of clinicopathological indices showed that AFP,preoperative GGT,themaximum diameter of the tumor,liver cirrhosis,differentiation and microscopic vascularinvasion are the risk factors of large hepatocellular carcinoma recurrence after radicalresection (P<0.05). Multivariate analysis showed AFP, tumor size is an independent riskfactor for recurrence within six months,while the microscopic vascular invasion is anindependent risk factor for recurrence within6~12months (P<0.05). 2. The expression of ALDH3A1in liver cancer tissues had significant difference in differentrecurrence time group (P<0.05). Moreover, the longer time of recurrence/metastasis inlarge HCC, the higher expression of ALDH3A1is. While the ALDH3A1gene expression inadjacent tissues showed no statistically significant (P>0.05).3. Multivariate analysis of ALDH3A1combined with clinicopathological factors showedthat the expression of ALDH3A1is an independent protective factor for recurrence andmetastasis within6months and6~12months; while independent risk factor for recurrencewithin6~12months is microscopic vascular invasion.Conclusions1. The expression of ALDH3A1in large HCC tissue is related with early recurrence andmetastasis, and with the longer time of recurrence/metastasis of large HCC, the higherexpression of ALDH3A1is.2. AFP,preoperative GGT,the maximum diameter of the tumor,liver cirrhosis,differentiation and microscopic vascular invasion are the risk factors of largehepatocellular carcinoma recurrence after radical resection;AFP, tumor size andmicroscopic vascular invasion are independent risk factors while the expression ofALDH3A1is an independent protective factor for recurrence and metastasis。... |
