| Objective:To evaluate the impact of choroidal neovascularization with macular hemorrhageclinical efficacy while Jiajian Zhujing Decocation, preliminary discuss the molecularmechanism of this method which inhibited choroidal neovascularization, fuether toshow objective evidence of Jiajian Zhujing Decocation treatment high myopia withchoroid neovascularization.Methods:1. Forty-two patients (42eyes) were divided into2groups: Jiajian ZhujingDecocation group(25eyes in twenty-five cases) treated with Jiajian ZhujingDecocation, and combined group administrated Jiajian Zhujing Decocation withintravitreal Bevacizumab injection on PM CNV(17eyes in seventeen cases). Centralretinal thickness were measured by optical coherence tomography prior to treatment.3month after the treatment, the therapeutic effect was observed.2. Rat choroidal vascular endothelial cells were cultured,(1) MTT assay wasused to detect CoCl2,Jiajian Zhujing Decocation containing serum and blank serumto stimulate the optimal concentration of rat choroidal endothelial cellproliferation.(2) We detect the effect of different treatments on Choroidal vascularendothelial cells secretion of HIF-Iα and VEGFmRNA by RT-PCR..Results:1. The total effective rate of combined group was up to82%3months after thetreatment, which of Jiajian Zhujing Decocation group was up to60%; in terms ofimproving retinal hemorrhage, combined total effective rate of88%after3monthsof treatment, the effective rate of addition and subtraction in Jing Fang group64%;in reducing central retinal thickness, each group after treatment compared withbefore treatment and two between the two groups, the differences were statisticallysignificant(p<0.05).2. The effect of CoCl2on the proliferation of Choroidalvascular endothelial cells was measured by MTT assay. The results showed the ODvalues of100umol/l were high than any other group. The difference between the twowas statistically significant(p<0.05). The results showed that the OD values of20% of jiajian Zhujing Decocation medicine serum were low than other group. Thedifference between the two was statistically significant(p<0.05). but The OD valuesof20%The OD values of Modified Zhu Jing Formula medicine serum wasstatistically significant than black serum group(p<0.05). The OD values of modelgroup,treatment group and control group compared with normal control group wasstatistically significant(p<0.05).The results of RT-PCR showed that modelgroup,treatment group and control group compared with normal control group,HIF-1αã€VEGF relative expressions was statistically significant(p<0.05).Conclusions:1. Jiajian Zhujing Decocation can delay the development process of high myopiamacular hemorrhage, at the same time improve the patient’s visionas well asimprovepatients′visual function.2. Jiajian Zhujing Decocation medicine serum couldinhibit hypoxia choroidal endothelial cells by inhibiting the proliferation of ratchoroidal endothelial cells secrete HIF-Iα, VEGF.3. Jiajian Zhujing Decocation mayhave to improve circulation, improve ischemia and hypoxia can regulate vascularendothelial cells secrete cytokines and other effects, which may be one of themechanisms of Jiajian Zhujing Decocation in the treatment of high myopia choroidalneovascularization. |
