Objective:Study of fibroblast proliferation and PCNA expression after matrinein glaucoma filtration surgery in rabbit eyes, Preliminary to explore anti-scaringeffect of matrine after glaucoma filtration surgery.Methods: Choose healthy New Zealand white rabbit40(male and femaleunlimited), only1.52.0Kg weight,these rabbit were randomly divided into fourgroups(each group including20eyes of10rabbits): group A group B group C groupD. Both eyes in four groups were treated with glaucoma filtration surgery. cottonpieces under the sclera flap were made:1.0g/l matrine cotton piece in group A,0.4g/l matrine cotton piece in group B,0.2mg/l mitomycin cotton piece in group C, ingroupD. Respectively to observe groups of intraocular pressure, filtering bleb,anterior chamber reaction, complications, and so on in the preoperative andpostoperative1,3,7,14,28days. Two rabbits selected randomly in every group werekilled on days1,3,7,14and28after surgery.Tissue was harvested from the bleb areaand was made pathological section, immunostained and Masson trichromatic dyeingTo observe the inflammatory cells, collagen, fibroblasts, the change of musclefibroblasts and new collagen fibers under light microscope. Two rabbits selectedrandomly in every group were killed on days7after surgery. Remove eye for temslice, to observe the toxicity of drugs.The data was analyzed statistically by SPSS17.0for windows.Results:1.IOP:Postoperative1,3days, four groups of IOP was not significantlydifferent (P>O.05), and postoperative7,14,28days,A, B, C group and D group twotwo comparison of IOP were significantly different(P<O.05), postoperative7,14,28days,A, B group had significantly different (P<O.05).2.Filtering blebs: Postoperative1,3days,each group of filtering blebs werediffuse,flat and transparent; postoperative14d, each group of filtering blebs werebigger than before, diffuse,ridgy and transparent. Postoperative7,14days A, B, Cthree groups of filtering blebs showed functional,and were diffuse,ridgy andtransparent,but group D of filtering blebs were flatter than before. 3. Inflammatory reaction: Postoperative1,3days,four groups appeared a smallamount of inflammatory cell infiltration, vascular dilatation, loose connective tissueedema and interstitial edema; postoperative7day, four groups showed massiveinfiltration of inflammatory cells, and small amount of new blood capillary dilate, andhyaline degeneration of connective tissue. postoperative14day, A, B, C three groupsof inflammatory cells were significantly reduced, there were only a small amount offibroblasts were,while in group D, there were still a large number of inflammatorycell infiltration, tissue structure of edema, more fibroblast deposition andangiogenesis; Postoperative28day, A, B, C three groups of only had a fewinflammatory cells, fibroblasts collagen and a small amount of fiber deposition, Dgroup had a large number of collagen fibers.Group inflammatory reaction in A and Cgroups was better than group B in each time point, D group was the most obviousinflammatory reaction.4.Fibroblasts were: Postoperative1,3days, fibroblast cell count of A, B, C threegroups showed no significantly different (P>O.05),compared with group D;Postoperative7,14,28days, fibroblast cell count of A, B, C three groups showed verysignificantly different (P<O.01),,compared with group D; And there wassignificantly different (P<O.05) between A and B. Besides there was nosignificantly different (P>O.05) between Aand C.5.Collagen changes: A, B, C groups of collagen fibers were less than that in Dgroup, there were large, dense,and arranged collagen fibers in group D.6.Transmission electron microscope: Corneal endothelial ultrastructure of groupC was damaged. to corneal endothelial ultrastructure, other groups were normal.Conclusion:(1)Matrine can relieve inflammation of filtration area afterglaucoma filtration surgery, inhibit fibroblasts hyperplasia, and reduce the scar offiltration area.(2)This study found matrine had no side effects of eye tissues.(3)Anti-scaring effect of1.0g/l matrine is better than0.4g/l matrine.
