ObjectiveTo study the effect of Eucommia leaf suppression adipogenic differentiation ofBMSCs and Screening of active substance.MethodsSD rats (about a month old)1, The SD rat BMSCs was separated by wholebone marrow paste the wall method; The cells which we have obtained weredetection by flow cytometry; Trypan blue staining and MTT assay cell growth curve;We formulated adipocyte differentiation inducing medium, Preliminary study leafextract and and its compounds, Experiment was divided into four groups: drugs(1μg/ml)+positive, the drug (0.01μg/ml)+positive,positive,negative group;Tofurther study on the active substance,Experiment was divided into five groups: drugs(1μg/ml)+positive group, the drug (0.1μg/ml)+positive group, the drug(0.01μg/ml)+positive, positive, negative group). drug intervention was terminatedafter14days,We continue to observe the morphological changes in eachexperimental group, and cells were staining and counted after oil red O staining;Todetected adipogenic differentiation PPARγ2and C/EBPα expression levels byquantitative PCR.ResultsFlow cytometry showed that CD29, CD90-positive rates were93.86%,92.16%,CD34,CD45-positive rates were6.99%,5.31%;rypan blue method counted3,4,5-generation BMSC.we found similar trends in different generations of cellgrowth, To test third-generation cell growth by MTT, the result is consistent with thetrypan blue counts; Oil Red O staining and counting fat cell differentiation rate,Eucommia leaf alcohol extract,quercetin,(+)-Syringaresinol each intervention groupless than positive group (p<0.05), and drugs have concentration-dependent manner;Quantitative PCR detected gene, results showed that:compared with the positivegroup, the PPARγ2and C/EBPα expression levels of each drug in the interventiongroup were reduced significantly different (p<0.05),and has a concentration-dependent manner.ConclusionEucommia leaf extract and its compounds quercetin,(+)-Syringaresinol havethe effect of suppression adipogenic differentiation,and has a concentration-dependentmanner, which is associated with PPARγ2and C/EBPα mRNA down-regulation ofthe expression levels.
