Study On Apoptosis Of Human Adenoid Cystic Carcinoma Cell Line ACC-M Induced By Garlic Oil Combined With Cisplatin

Objective:To investigate the effect of garlic oil combined with cisplatin on humanadenoid cystic carcinoma cell’s proliferation,cell cycle and apoptosis in the hope ofofferring a theoretical basis and experimental evidence on combination therapy of garlicoil and cisplatin in prevention and treatment of this cancer.Methods:The experiment was divided into groups of garlic oil,cisplatin and garlic oilcombined with cisplatin(1:1)and no drug treatment in ACC-M cells as the controlgroup.ACC-M cells were cultured with garlic oil,cisplatin,garlic oil combined cisplatin indifferent time(4,8,12h) and then detected the inhibition of cells by MTT assay.Flowcytometry was used to analyse of the changes of cell cycle and apoptosis rate after ACC-Mcells were treated with garlic oil,cisplatin,garlic oil combined cisplatin as long as4,8h.Results:①Each drug treatment group had inhibition on ACC-M cells.With drugconcentration increased,the inhibition became more and more obvious(P<0.01).32μg/mLgarlic oil combined with cisplatin cultured with ACC-M cells in12h was the bestcombination,the inhibition rate was77.16%. The garlic oil and cisplatin combinationtherapy suppress the growth of ACC-M cells was significantly stronger than a singletreatment group(P<0.01).②garlic oil,cisplatin,garlic oil combined with cisplatin couldchange the cell cycle distribution of ACC-M:with the increase of concentration,the G0/G1phase of the cell rate decreased,S phase had no significant change,but G2/M phase cellsincreased(P<0.01).Compared with the single drug group,32μg/mL garlic oil combinedwith cisplatin cultured with ACC-M cells in8h made the G2/M phase arrested moresignificant (P<0.01).③Flow cytometry showed that garlic oil,cisplatin,garlic oil combinedwith cisplatin could induce apoptosis of ACC-M cells.With the increase of concentration,the apoptosis rate rose,the apoptosis rate of32μg/mL garlic oil combined with cisplatin cultured with ACC-M cells in8h was7.64%.The role of combination therapy group wassignificantly stronger than the single drug group(P <0.01).Conclusion:①Different concentrations of garlic oil,cisplatin,garlic oil combined withcisplatin all have significantly growth inhibition on human adenoid cystic carcinomaACC-M cells,32μg/mL garlic oil combined with cisplatin cultured with ACC-M cells in12h was the best combination.②2、8、32μg/mL garlic oil,cisplatin,garlic oil combined withcisplatin could induce ACC-M cells apoptosis and arrest cell cycle in G2/M phase.In acertain range,there is a dose dependent manner,The effect of32μg/mL garlic oil combinedwith cisplatin cultured with ACC-M cells in8h was more obvious.③The effect of garlicoil combined with cisplatin on ACC-M cells was stronger than the garlic oil,cisplatin usedalone.
