Objective:To investigate the effects of periostracum cicadae and bombyx batryticatus on the treatment of mesangial proliferative glomerulonephritis(MsPGN) of Rats, to explore its mechanism and compare the difference between periostracum cicadae and bombyx batryticatus.Methods:A total of70Wistar rats in clean grade,weighing200g±20g, were adaptivcly fed in the laboratory for a week.10rats were randomly divided into normal control group. And the models of MsPGN rats were established by ameliorated chronic disease serum methods. Five weeks later, Coomassie brilliant blue method was used to detected24h urinary protein. The rats with negative24h urinary protein were excluded. Then the models were randomly divided into model control group, low periostracum cicadae group[1.75g/(kg.d)], high periostracum cicadae group[3.5g/(kg.d)], low bombyx batryticatus group[1.75g/(kg.d)], high bombyx batryticatus group[3.5g/(kg.d)].Each group had10rats. After the administration of5w, all rats were detected on24h urinary protein. Eight weeks later, all rats were detected on24h urinary protein. Blood biochemical indicators were detected, and pathological changes were observed by hematoxylin-eosin staining(HE),and the expression levels of TLR4and TGF-131were detected by immunohistological method.Results:After the administration of5w, compared with the model control group,24h urine protein decreased in both high dose groups, and the differences were significant differences (P<0.01); After the administration of8w,24h urine protein decreased significantly in each dose groups. Compared with the normal control group, ALB decreased significantly, TC and BUN increased significantly in the model control group. Compared with the model control group, TC decreased significantly in every dose groups; ALB increased significantly in periostracum cicadae groups; BUN decreased significantly in the high periostracum cicadae group and the differences were significant (P<0.01). Morphological observation of the renal tissue showed that in both high dose groups glomerular mesongial cells in individuals regions had mild hyperplasia, and mesangial area slightly widened, and there was no extrusion in lumen, Significant improvement compared with the model group. Compared with the model control group, the distribution of TLR4in kidney tissue cytoplasm decreased significantly in each dose groups, and the differences were significant differences (P<0.01), and there were no significant differences in periostracum cicadae and bombyx batryticatus groups. Compared with the model control group, the distribution of TGF-β1in kidney tissue cytoplasm decreased significantly in each dose groups, and the differences were significant differences (P<0.01),and there were no significant differences in periostracum cicadae and bombyx batryticatus groups.Conclusion:Periostracum cicadae and bombyx batryticatus may decrease24h urinary protein, ruduce cholesterol on MsPGN rats, and improved the pathological changes in rat kidney, its mechanism may be related to inhibiting over-expressiom of TLR4and TGF-β1.
