Objective The aim of this study was to determine the association of interleukin-1(IL-1)gene promoter and interleukin-6(IL-6) gene promoter polymorphisms with the chronicperiodontitis in Hui and Han populations from Ningxia area, and to analyze the distributionsof genotypes and allele frequencies of IL-1gene promoter polymorphism and IL-6genepromoter polymorphism between the two nationalities.Methods We collected peripheral venous blood from participants, including132patients with chronic periodontitis (CP) and138healthy controls in Hui nationality;141patients with chronic periodontitis (CP) and146healthy controls in Han nationality. In ourstudy, the SNPs of IL-1B-511(rs16944) and IL-6-572(rs1800796) were investigated bypolymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)respectively. We evaluated the genotypes and alleles of IL-1B-511and IL-6-572from all thesubjects and analyzed that whether there are associations of IL-1B-511and IL-6-572genepolymorphisms with CP by case-control methods.Results (1) IL-1B-511(rs16944): In this study, we found2alleles (allele C and T)and3genotypes (genotype C/C, C/T, T/T). There was statistically significant difference in thefrequencies of IL-1B-511genotypes and alleles between the CP patients and healthy controlsin Hui nationality, and the frequency of the allele T in the CP group is higher than in thehealthy controls (P=0.027and P=0.018, respectively); There was statistically significant difference in the frequencies of IL-1B-511genotypes and alleles between the CP patients andhealthy controls in Han nationality, and the frequency of the allele T in the CP group is higherthan in the healthy controls (P=0.034and P=0.037, respectively); There was no statisticallysignificant difference in the frequencies of IL-1B-511genotypes and alleles between CP ofHui nationality and CP of Han nationality (P=0.908and P=0.641, respectively);(2)IL-6-572(rs1800796): In the study, We found2alleles (allele C and G) and3genotypes(genotype CC, CG and GG). There was no statistically significant difference in thefrequencies of IL-6-572genotypes and alleles between CP patients and healthy controls inHui nationality (P=0.561and P=0.321, respectively); there was no statistically significantdifference in the frequencies of IL-6-572genotypes and alleles between CP patients andhealthy controls in Han nationality (P=0.731and P=0.413, respectively); there was also nostatistically significant difference in the frequencies of IL-6-572genotypes and allelesbetween the Hui patients with CP and Han patients with CP (P=0.878and P=0.599,respectively).Conclusion (1) IL-1B-511C/T (rs16944): The polymorphism of the IL-1B-511wasassociated with the chronic periodontitis in Hui nationality and Han nationality, but there wasno significant difference between Hui and Han ethnicity.(2) IL-6-572C/G (rs1800796): Thepolymorphism of the IL-6-572was not associated with the CP in Hui and Han nationality andthere was no difference between the two ethnicities.
