Object Detection of hepatocellular carcinoma (HCC) in Aflatoxin B1-DNA adduct levels of exposure and cyclooxygenase2(COX-2), the expression of p53protein, in combination with the clinical pathological data, analysis of the three in the HCC and non HCC expression level differences, of three of HCC effects of and between the three mutual correlation and clinical significance.Method Collected in our hospital since2006January to2010September operation excision of50cases of patients with liver cancer and40cases of patients with hepatocellular carcinoma liver tissue paraffin block specimens. Using immunohistochemical staining method for the detection of the specimens of AFB1-DNA adducts, cyclooxygenase2(COX-2), expression of p53protein, analyzes three and clinical pathological factors mutual association.Result1.AFB1-DNA adducts in HCC positive results in localized in the nucleus. HCC AFB1-DNA adducts in positive rate was significantly higher in carcinoma and non HCC liver tissue, the difference was statistically significant (P<0.05). AFB1-DNA adduct levels of exposure and the serum alpha-fetoprotein (AFP) were significantly correlated (P=0.014), liver cancer and the TNM staging (P=0.502), the size of the tumor (P=0.478), histological grade (P=0.083) and liver metastasis (P=0.560) with no obvious correlation.2.COX-2in primary hepatocellular carcinoma tissue expression was localized in the liver cytosol, brown yellow granular. HCC COX-2positive rate was significantly higher than that of and the size of the tumor (P=0.478), TNM staging (P=0.791), AFP (P=0.393), extrahepatic metastasis (P=0.560) and other factors had no significant correlation (P>0.05).3. p53protein positive expression of HCC in cancer cells, brown yellow granular. p53positive rate was significantly higher in HCC than non HCC liver tissue, the difference was statistically significant(P<0.05).P53protein surface expression level and pathological grading (P=0.046), extra-hepatic metastasis (P=0.014) were significantly related to liver cancer,and TNM stage (P=0.053),the size of the tumor (P=1.000) and serum AFP(P=0.981) and no obvious correlation. AFB1-DNA adduct levels and the expression of p53protein levels were positively correlated with p53, COX-2expression levels are positively correlated, AFB1-DNA adduct levels and COX-2expression level was not significantly related to.Conclusions AFB1is one of important factors in the development of hepatocellular carcinoma. Intrahepatic COX-2high level expression of further enhancing the AFB1carcinogenic toxicity. AFB1and COX-2alone or in combination to p53enhanced protein expression. AFB1, COX-2and p53proteins in HCC occurred, which existed in the development of cooperative relations, analysis of three relations for further study of HCC carcinogenic mechanism has a certain reference value. |
