Objective: Compare the sensitivity of the eight recombinant antigens suchas Zw1ã€Zw5ã€Zw14ã€14-3ã€Zw15ã€Eg10ã€P29ã€Hsp70to investigate the diagnosticvalue of the eight recombinant antigens of liver Echinococcus granulosusdisease.Methods:1.Samples prepared: we collect the serum of53patients of liverEchinococcus granulosus diagnosed by surgery and60nomal serum;2.concentrationof the recombinant antigen evaluated: we get the concentration of the recombinantantigens by the application of BCA protein quantitative method;3.antigen purityidentification: we get the clear size of the recombinant antigen molecular weightand analysis the bands on the gel to identify the antigen purity by the imageof SDS-PAGE on the gel imager;4.identify the diagnostic value of the recombinantantigen: we get a better diagnostic value antigen of liver Echinococcusgranulosus disease which has a higher sensitivity in the results of ELISA.Results:1. On the gel imager, the size of the eight recombinant antigensweight is between18-90KDa, the gel bands of the eight recombinant antigens areclear, the recombinant antigens have high purity.2. BCA protein quantitativemethod for the determination or the concentration for the recombinant antigensdefined the coating buffer concentration of microtiter plates,3.By ELISA,the zw14recombinant antigen and the Eg10recombinant antigen have higher sensitivity.Conclusion: The Zw14recombinant antigen and the Eg10recombinant antigenare expected to become the better diagnostic value antigens of liver Echinococcusgranulosus disease. |
