At present, atherosclerosis disease is one of most critical diseasesthat threaten human health. In western countries,coronary heart diseaseis the primary cause of death. Ross shown that inflammation plays anessential role in pathogenesis of atherosclerosis disease. So manystudies demonstrate that secretory phospholipase A2is a real player inthe development of cardiovascular diseases.0bjectiveThe study is to evaluate the effects of irbesartan on group V secretoryphospholipase A2(sPLA2-V) expression in aorta of rats withatherosclerosis,and explores the molecular mechanism furthermore.MethodsThirty healthy male Wistar rats were equally randomized into threegroups(n=10each):control group,model group,and irbesartan group. Thecontrol rats were fed with normal diets,the other rats of model groupand irbesartan group(25mg·kg-1·d-1pergavage) were fed with highcholesterol diet.After12weeks, the expression of group V sPLA2wasmeasured by immunocytochemistry and the gene expression of group V sPLA2was detected by reverse transcription polymerase chain reaction(RT-PCR)Results①sPLA2-V was present in smooth muscle cell and the plaque of aorta. Incontrast, the expression of sPLA2-V in aorta was much higher in modelgroup than in controls(P<0.01).The expression of sPLA2-V in aorta wassignificantly decreased in irbesartan group compared to model group(P<0.01).②Compared with control group, the expression of sPLA2-V at protein andmRNA level of the model group was significantly elevated (P<0.01).The activity could be attenuated by irbesartan intervention (P<0.05vs modelgroup).Conclusions The elevation of sPLA2-V is binding to the pathogenesis ofatherosclerosis,which may be a player in atherogenesis. To acertain extent,irbesartan could relieve the process of atherogenesis bydecreasing the expression of group V sPLA2in aorta.
