The Effect Of Bupi Yizhi Method On The Ability Of Learning And Memory And The Expression Of NGF, BDNF,NT-3 In Rats With Alzheimer’s Disease

Objective:To observe the effect of Bupi Yizhi Method on the ability of learning and memory and the expression of NGF,BDNF,NT-3 in rats with Alzheimer’s disease(AD),and to explore the therapeutical effect and the potential mechanism, in order to assistant the treatment of AD with integrated Traditional Chinese Medicine.Method:Fifty-two SD rats were classified into 5 groups at random:a normal group, a sham-operated group, a model group, a Traditional Chinese Medicine group(Bupi Yizhi Method group), and a Western Medicine group(Peileineng group).The Alzheimer’s disease model was established by Aβ1-40 injected into the hippocampal of the rats. The ability of learning and memory was evaluated by Morris water maze and the Shuttle-box.The expression of NGF, BDNF, NT-3 was detected with immunohistochemical staining.Results:1 After operation, compared with the normal group, the Traditional Chinese Miedicine group,the Western Medicine group and the model group had a longer platform searching latency(P< 0.01),less times of passing through the original platform position(P< 0.01),more shocked times(P<0.01) and longer shock time(P<0.05), the sham operation group had no significantly different with the normal group(P>0.05);Compared with the model group,there was no significant difference in the Traditional Chinese Medicine group and the Western Medicine group(P> 0.05).2 After treatment, compared with the model group, the Traditional Chinese Medicine group,the Western Medicine group had a shorter platform searching latency(P< 0.01), more times of passing through the original platform position(P< 0.01),less shocked times(P< 0.05) and shorter shock time (P<0.05);The traditional Chinese Medicine group had no significant difference with the Western Medicine group (P>0.05).3 By contrasting the evaluation of learning and memory ability after treatment with before in the Traditional Chinese Medicine group,after treatment,the model group had a shorter platform searching latency(P< 0.05),more times of passing through the original platform position(P< 0.01),less shocked times(P<0.01) and shorter shock time(P<0.01) than before.4 Detecting with immunohistochemical staining, compared with the normal group, the expression of NGF in the hippocampus of the model group was significantly increased (P<0.01); Compared with the model group, the expression of the NGF in the Traditional Chinese Medicine and the Western Medicine group was significantly increased (P<0.01);Compared with the Western Medicine group,there was no significantdifference on the expression of NGF in the Traditional Chinese Medicine(P>0.05).5 Detecting with immunohistochemical staining, compared with the normal group, the expression of BDNF in the hippocampus of the model group was significantly decreased (P< 0.01); Compared with the model group, the level of the BDNF in the traditional Chinese Medicine was significantly increased (P<0.01),while that in the Western Medicine group was also significantly increased (P<0.05);Compared with the Western Medicine group,there was no significant difference on the expression of BDNF in the Traditional Chinese Medicine(P>0.05).6 Detecting with immunohistochemical staining, compared with the normal group,the expression of NT-3 in the hippocampus of the model group has no significant difference (P>0.05);Compared with the model group, the expression of the BDNF in the Traditional Chinese Medicine was significantly increased (P<0.05), while that in the Western Medicine group was also significantly increased (P<0.01);Compared with the Western Medicine group,there was no significant difference on the expression of BDNF in the Traditional Chinese Medicine(P>0.05).Conclusion:1 The Alzheimer’s disease model was successfully established by Aβ1-40 injected into the hippocampal in the rats.2 Bupi Yizhi Method can distinctly improve the learning and memory ability of rats with Alzheimer’s disease, it has obvious therapeutical effect on Alzheimer’s disease.3 After treatment of Bupi Yizhi Method, the expression of NGF,BDNF,NT-3 incresed in the rats with Alzheimer’s disease, which suggest that these three factors paly important roles in the therapeutic process,the function mechanism mainly is that Bupi Yizhi Method can increase the nutrition support for nervous system.4 Bupi Yizhi Method can improve the learning and memory ability of rats with Alzheimer’s disease and increase the expression of NGF,BDNF,NT-3,which proved that it can be used to treat AD,and it also furnish evidence to Congpi Lunzhi treatment on Alzheimer’s disease.
