Abstract:Objective:Euodiae fructus is a commonly used traditional Chinense medicine. The quality of Evodia from different areas are different. For a long time, because of the usage of different evaluating methods and standards, people have different comments on the quality of Euodiae fructus from different areas. Because of the complex components of traditional Chinese medicine, a single active ingredient analysis can not represent its clinical effects. Therefore we can not only use a single active ingredient as a standard to evaluate Evodia. So in this experiment, HPLC method was used to determine the several active ingredients and ISSR was used to compare the genetic similarity of the Evodia from different areas.Methods:(1)The Morphology methods were used to identify different kinds of Evodia.(2)The volatile oil contents of Evodia form different areas are analysed,too. (3)HPLC method was used to determine the major components of Evodia. (4)ISSR is used to compare the genetic similarity of different kinds of Evodia from Sichuan and Chongqing. Results:(1)Evodia from Sichuan and Chongqing has three species: Evodia rutaecarpa (Juss.) Benth., E. rutaecarpa (Juss.) Benth. var. bodinieri(Dode)Huang and E. rutaecarpa (Juss.) Benth. var. officinalis (Dode) Huang. The three species can be distinguished from their form/microscopic and physical characteristics.(2) Although all the three species of Evodia are rich in volatile oil, but their contents are related to the areas of origin/picking time/storage time and storage conditions. The volatile oil contents of the three species of Evodia have no significant difference. On the other hand, there is no volatile oil in their stalk. (3) The limonin contents in 21 samples are too low and do not meet the requirements of pharmacopoeia. (4)ISSR method was used to analyse the genetic similarity of 59 Evodia samples from Sichuan and Chongqing.20 primers which have good amplification effect and stability are selected to constructed the ISSR map. The genetic similarity between different kinds are compared and the dendrogram map of Evodia was constructed, too. We altogether amplified 132 strips, each primer could amplify 5 to 9 strips, the average is 6.6,105(80.5%) strips have polymorphism. At the similarity level of 0.83,the 59 Evodia samples can be divided into 7 categories. We can find the significant difference of genetic distance between Evodia from different areas, therefore the genetic variation is tightly related with the geographical distribution. Conclusion:This article focused on the usage of quality indicators(color/plumpness/oder) and physical and chemical analysis(volatile oil content/the total alkaloid content) to evaluate the Euodiae fructus quality.This experiment combined contents and components,quality intergrating with genetic similarity to evalute the quality of Euodiae fructus.From the results of the morphology/volatile oil/alkaloids/limonin,we can draw the conclusion that the quality of Evodia from Chongqing Qianjiang is better than other areas. The ISSR results showed that Evodia from Chongqing Youyang banxi, Chongqing Hechuan and Chongqing Qianjiang zhaishui have higher genetic similarity, they have close genetic relationship.Therefore we can draw the conclusion that the quality of Evodia from Youyangbanxi and Hechuan are better than others. We established a method to evaluate the quality of Euodiae fructus.
